The heart development is a very complicated process which is controlled by a series of highly conserved transcription factors (such as NK2, MEF2, GATA, Tbx, and Hand). These transcription factors decide fates of cadiocytes, expressions of genes coding for spectrin and morphogenesis of the heart At the same time, they also play critical roles in the regulation of neurogenesis, cadiocytogenesis, hemopoiesis and cancer cytohistogenesis.In this study we put on researches on gene Yippee. It encodes a highly conserved zinc finger binding protein, and its homologous protein was found in 68 kinds of eucaryon organisms from eumycete to human. The expression of Yippee extensive in different tissues of human embryos and in different development stages of Drosophila indicates it may be involved in important biological processes. In 2003, the proteins interaction map of Drosophila was accomplished and the result showed that Yippee may regulate heart development related genes such as Wg, Mef2 by interacting with CG15676, suggesting Yippee may be probably involved in signal pathways related to heart development. So we presume that Yippee may have functions during heart development. We obtained Drosophila Yippee cDNA by using RT-PCR and constructed green fluorescence overexpression vector pUAS-Yippee-IRES-GFP containing Yippee and IRES-GFP. Successfully we obtained Yippee transgenic flies through Drosophila embryo microinjection and mini-white mark gene screening. Then we crossed the Yippee transgenic fly line with Hand-GAL4 fly line and RNA interference Yippee fly line with Hand-GFP fly line respectively, and watched the phenotypes of the filial generation under a fluorescence microscope so as to determine the potential roles of Yippee in regulating heart development.
