With the development of genome-wide sequencing technology, more and more noncoding RNA (ncRNAs) were characterized, and their cellular roles are in the process of being revealed. Now ncRNA have become a hotspot in the life science. These small molecules exist in almost all higher eukaryotes, they lack of clearly identifiable open reading frame (ORF) to translate functional proteins directly,and have very important regulation roles in these organisms. In the previous work, our laboratory found a noncoding gene like mRNA by suppression subtractive hybridization that called Z38. The gene is highly expressed in breast cancer,and there is no report on the gene's function.In order to study biological functions of Z38 in vivo, this study constructed a eukaryotic expression vector of Z38 that plus the stop codon, then transfected HBL100 cells and injected in nude mice. The results showed that three mice had generated tumors in five nude mice,but in the control group, only one nude mouse had generate a tumor that growed lately and slowly.At the same time, we constructed a eukaryotic expression vector for Z38,in which the expression of Z38 is driven by a MMTV promoter,transgenic mice for Z38 were generated by microinjection of MMTV-EGFP-Z38,four fouder mice were obtained for Z38 after PCR analysis,and the four mice had undergone breast tumors, after HE staining proved to be breast carcinoma. Extraction of the breast tumor tissues RNA of these transgenic mice, we demonstrated the high expression of Z38 in these tumor tissues by PCR.In summary,we successfully constructed the transgenic mice of nocoding RNA Z38,and Z38 is highly expressed in breast tumor tissues of transgenic mice. The establishment of the transgenic mice provides us a model for studying the function of Z38 in vivo.
