Ginseng is a kind of precious traditional Chinese herbal medicine. It possesses cardiotonic and hypotensive effects, promotes immune function and metabolism, anti-stress and anti-aging activities. Nowadays, people pay more and more attentions to medicinal value of ginseng, so the market demand of ginseng increase gradually. However, on the market, there was mainly artificially cultivated ginseng. To increase their high yields, some of the organochlorine pesticides and organophosphorus pesticides such as pentachloronitrobenzene (PCNB), and diazinon are widely used as antiseptic. They possess high toxic and bioaccumulation; which are eventually hazardous to human health through the food chains. Therefore, fast determination of residue pesticides from ginseng is necessary to protect human health. Ginseng is a complex matrices contained large numbers of spooning, polypeptides and saccharides, which removed from the crude extract due to interference in gas chromatographic analysis. Most of general sample pretreatment methods for ginseng samples were including extraction, purification and concentration processes. Up to now, the most common method is concentrated sulfuric acid sulfonation purification (CSASP), While, the methods are time-consuming, require large volume of toxic organic solvents, and difficult to automate. In addition to some of pesticides are unstable under the acidic condition. To overcome these shortcomings, our laboratory developed a "gas purge microsyringe extraction" (GP-MSE), All the extraction and clean up process were occurred in a 250μL microsyringe. This study established a gas purge microsyringe extraction coupled with gas chromatography-mass spectrometry detection of organochlorine pesticides and organic phosphorus pesticide (including Pentachloronitrobenzene and its metabolites Pentachloroaniline, Pentachlorothioanisole, Phorate, Dichlorvos, Diazinon, Omethoate, Dimethoate, Malathion, Quinalphos, Triazophos,) in ginseng. Based on previous publication, kinds of extracting solvent and extraction temperatures were improved in here, and added average recovery was 78.1-108.6% and relative standard deviation was less than 9.8%. Compared with Soxhelt extraction and concentrated sulfuric acid sulfonation purification, this method is faster, simpler, greatly reducing the amount of organic solvent and sample, high sensitivity, good reproducibility and accurate qualitative and quantitative analysis, while application of this method for measuring other herbs and pesticides residues in crops has a good prospects.
