| C60 is different from the ordinary aromatic benzene ring compounds, its unique structural features , excellent physical and chemical properties made them have great potential applications in fields of chemistry, physics, materials ,biotechnology, optoelectronics , life science,medicine and other areas of academic research. In this paper, according to the molecular design principle and using a variety of synthetic methods, a series of new C60 derivatives were synthesized for the first time. They are C60-glutamine, C60-serine, C60-proline derivatives, C60-malonyl dihydroxy amines and C60-curcumin.These new C60 derivatives were also characterized by some analytical tools. Meanwhile, we used chemiluminescence technique to detect these C60 derivatives on hydroxyl radical scavenging efficiency. In addition , the distribution in vivo of two water-soluble C60 derivatives which have been synthesized previously were discussed. The main contents include:(1). C60-glutamine, C60-serine, C60-proline derivatives, C60-malonyl dihydroxy amines and C60-curcumin have been synthesized, and we also studied different factors which influenced the synthesis. The optimal reaction conditions are as following: time 16h, the molar ratio of glutamine and C60 200:1, and the amount of catalyst (10%)tetrabutyl ammonium hydroxide (TBAH) 400μL.(2).The new C60 derivatives are characterized by UV spectrophotometer (UV-Vis), Fluorescence spectrophotometer (RF) , Fourier transform infrared spectroscopy(FT-IR), nuclear magnetic resonance NMR(1HNMR),carbon NMR (13CNMR),elemental analyzer, X-ray diffraction(XRD) and other methods.(3).We used o-phenathroline chemiluminescent system and chemiluminescence to detect hydroxyl radical scavenging efficiency. Compare the four C60 derivatives in the 800μg/ml concentration, the scavenging activity was with the following sequence: C60-glutamine 99.2%> C60-serine 71.8% > C60-curcumin 71.2%> C60-malonyl dihydroxy amines 70.8%. The impact of C60 derivatives on hydroxyl radical scavenging were also been analyzed.(4). The water-soluble biocompatible C60-phenylaanine, C60-βalanine were selected as objects of experiment of distribution in vivo. According to the experimental results we also discussed various possible analyses. |
