Micro/Nano-controlled drug delivery system has been a hot research field of controlled release.Chitosan has excellent biocompatibility and biodegradability. Chitosan as drug carriers have been made extensive and in-depth study.In this thesis,the chitosan microspheres were compared by emulsion solidification method and ionic cross-linking method. The nanoscale chitosan microspheres were prepared by Single coagulation method. The urokinase-loaded microspheres were prepared by adsorption method. SEM,TEM and Laser particle size analyses were investigated to character the microspheres morphology and particle size distribution.In this thesis,the drug-loaded microspheres conditions were discussed.The result of the experiment and direction for improvement were analyzed.The effect of urokinase and drug-loaded microspheres were determined by in vitro simulation of thrombolytic test.Results showed that chitosan microspheres containing urokinase can show good thrombolytic capacity. In this field chitosan microspheres showed application potential. This paper may provide a reference for further precise construction of drug-loaed chitosan microspheres thrombolysis system.
