Idebenone (IDBN) is an effective and safe natural antioxidant which can be used to preventskin from oxidative damage in surrounding environment, and prevent the development ofdesalinate fine lines, wrinkles and age spots or other related problems. Making IDBN intomicrocapsules can improve its water solubility and dilute the color of IDBN. Besides, theinherent physiological effect of IDBN can be maintained. In this study, we adopted β-cyclodextrin coating and spray drying to produce IDBNmicrocapsules. β-cyclodextrin coating method selected the factors which have a greater effect oncoating for orthogonal experiment based on single factor expriments. We have selected optimalmicroencapsulation parameter through evaluation of coating rate, yield, drug content and otherindicators. Spray drying method selected two sets of wall material: gelatin-maltodextrin andsodium alginate-gelatin to experiment. Then, the factors which had a greater effect on rate ofcoating were optimized, such as the emulsification temperature, wall material rate, homogenization time to get the optimum technology of preparing microcapsules. In the experiment of β-cyclodextrin coating, the optimum conditions of orthogonalexperiment are as follows: the molar ratio of IDBN and β-CD is1:3, the speed of electric mixer is400rpm,the reaction temperature is50℃, the time of ultrasonic inclusion reaction is20min, thecontain quantity of IDBN-β-CD can be achieved to14.77%, coating rate is35.20%, yield is30.08%.In the experiment of spray drying, based on the rate of coating, two sets of wall materialwere selected to prepare IDBN microcapsules. It shows that the concentration of sodiumalginate1%and the concentration of gelatin2.5%composite as wall material obtained a highercoating rate. The optimum conditions are as follows: the core-wall ratio1:2, homogeneousemulsion5min. The coating rates up to87.7%.X-ray diffraction, infrared spectroscopy and scanning electron microscopy were used totestify the formation of microcapsules. It shows that the orange appearance of IDBN is greatlydiluted after the IDBN has been made into microcapsule by these two methods. The solubility ofthe IDBN increased by70times after β-CD coated and increased by95times after coated bysodium alginate and gelatins. The study of IDBN microencapsulation in this paper not only expands the scope ofapplication of microcapsules, but also broadens the research thought to develop more new-typeand efficient cosmetic additives.
