In this paper, on the basis of existing research work, at first, applied in response surface method of S-adenosine-L-methionine optimized fermentation, then in 10L bioreactor in developing the SAM fermentation technology optimization research, in 50L and 600L reactor investigated SAM fermentation of stability and amplification feasibility, eventually fermentation technology successfully amplified to pick 6000L pilot, main conclusions include:1. The application Burman experiment, Plackett-the steepest hill-climbing experiment and response surface methodology to optimized based fermentation, get the optimal fermentation formula This formula (g/L) for:glucose 13.9 g/L, yeast extract 13.8 g/L, (NH4) 2SO4 8 g/L, K2HPO4 4.5 g/L, MnSO4 0.08 g/L, ZnSO4 0.8 g/L, MgCl2 0.1 g/L, CaCl2 1 g/L, FeSO4·7H2O 0.4 g/L, Met 23.3 g/L, In this formula, SAM content is 6.14 g/L, and optimize based fermentation before SAM content 2.65 g/L2.10L cans of S.cerevisiae fermentation research shows that add glucose concentration of dry weight and product of cells have significant effect on the accumulation of add glucose concentration, best for 45g/L; When fermentation with 2 g/(L·h) rate flow plus D-methionine precursor, SAM accumulation quantity is 1.53 g/L, biomass at 142 g/L; At the same rate flow plus precursor L-methionine, SAM accumulation quantity of 606 g/L, biomass for 147 g/L. In comparison, with 4 g/(L·h) rate flow plus DL-methionine precursor, SAM accumulation quantity, can reach the highest 2007 g/L, biomass for 145 g/L; Ethanol hand can control the bacteria than growth rate, on the other hand can promote intracellular synthesis of SAM synthase department, ethanol optimal concentration for 15g/L; Control pH 5.0, dissolved oxygen is the most suitable to 30% in SAM's synthetic biology.3. 50L bioreactor in fermented for 48h, mycelium concentration 142.8 g/L, SAM concentration for 6.18 g/L, In 10L,50L,600L bioreactor of the experimental results show that the process is easy to enlarge and good stability.4. SAM fermentation technology successfully amplified to 6000L tank, fermentation 48h, bacteria 143.3 g/L concentration, SAM concentration 6.26 g/L for.
