In this paper, the Cu(II) species were measured by diffusive gradients in thin films (DGT) technique, ion selective electrode (ISE) and dialysis peeper (Dialysis), respectively. The Cu(II) species existed in the synthetic solution at the different ratios (1:1,1:2,1:5 and 1:10) of ligands (FA and HA) to Cu2+ were assessed by DGT. The Cu(II) species existed in spiked river water, spiked domestic waste water and spiked industrial water were systematically investigated by the three motheds. In the Cu-FA system, the recoveries of DGT and ISE decreased with the increase of FA concentrations and the recovery of Dialysis decreased with the increase of FA, until to be a constant. In the Cu-HA system, the recoveries of DGT and Dialysis decreased slowly with the increase of HA concentrations, while the recoveries of DGT and Dialysis decreased sharply at the ratio of Cu:HA= 1:10. The recoveries of ISE decreased with the increase of HA concentrations and the concentrations of ISE-labile Cu were lower than the detection limit of ISE at the ratio of Cu:HA= 1:5 and 1:10. When the ratio of Cu:HA was less than 1:5,28 %of soluble Cu-HA complex was the DGT-labile species. The content of soluble DGT-inert Cu-HA complex decreased with the increase of HA concentration. The ratio of soluble DGT-labile Cu species to total soluble Cu species in water increased with the increase of HA.The Cu(II) species existed in spiked river water, spiked domestic wastewater and spiked industrial wastewater were systematically studied by the three methods. The recoveries of DGT for Cu2+in spiked river water, spiked live water and spiked industrial water were 9.920 %,7.122% and 6.671%, respectively; The recoveries of ISE in above three waters were 6.311%,3.190% and 2.110%, respectively; The recoveries of Dialysis in above three waters were 89.10%,76.36% and 42.83%, respectively. The DGT- and ISE-labile Cu(II) species were affected by the mount of dissolved organic carbon (DOC) in the three waters and the recoveries of DGT and ISE decreased with the increase of DOC concentration. In the three waters, about 4% complexes were DGT- labile Cu(II) species. The recoveries of Dialysis were affected by the mount, the species and the characteristic of DOC. For a method, the recovery of Cu2+in the three waters was not the same, it was due to the amount and species of natural organic ligands. The recoveries of the three methods for Cu2+ in the same water was also not the same, it was because the labile Cu (II) species of the three methods was different. The Cu(II) species could be measured by combination of DGT, ISE and Dialysis.
