Zwitterionic stationary phases have been widely used in separating and detecting inorganic ions in Ion Chromatography. Simultaneously, this kind of material also performs well in separating highly polar and hydrophilic compounds with high efficiency and selectivity. As we all know, lysine has zwitterionic structure in nature, and using it as ligand to modify stationary phases often showed great selectivity and efficiency in Ion Chromatography, however, there are relatively rare references reporting its application in separating hydrophilic and much polar compounds such as cephalosporins and carbapenems. Click Chemistry has many advantages such as high efficiency, high selectivity, and no special requirements to the reacting temperature and solvent, and compared with materials synthesized by traditional methods, stationary phases prepared through Click Chemistry often demonstrated good chromatographic properties.This research project prepared lysine functionized zwitterionic stationary phase (CLSP) utilizing the 1,3-dipolar cycloaddition reaction (CuAAC) between terminal alkynyl silica and azido derivative catalyzed by Cu (Ⅰ). Compared with traditionally prepared lysine zwitterionic stationary phase (TLSP), this new material showed better performance in Ion Chromtography, especially in analyzing anions with higher efficiency and selectivity, as well as good stability. More importantly, it could simultaneously separate various anions efficiently in a short time.CLSP also demonstrated great potential in separting hydrophilic and polar compounds such as nucleosides, aromatic acids and penicillins in Hydrophilic Interaction Liquid Chromatography (HILIC) mode with higher selectivity and efficiency in contrast to TLSP and commercially available Atlantics HILIC Silica or ZIC-HILIC column.
