China is the bamboo shoots export power, the annual output is3.2million tons.The shoots root and shell always be considered as bamboo shoots residue in factoriesprocessing, accounting for50~70%, and lots of them have been chopped and burned inthe open field by human being, which will cause ecological and environmentalproblems, and utter waste the resources. Therefore, it is imminent and economical tofurther processing the bamboo shoots residue, develop microorganism resources fully,and using the microorganism fermentation bamboo shoots residue conversion intophytosterol. In addtition, it could improve the economic benefits for bamboo shootsprocessing industry.Therefore, this study took ChongQing fang bamboo shoots residue as raw material.Through separated decay residues of bamboo shoot, got9kinds of strains, thenscreening the most powerful of producing phytosterol, obtained an Aspergillus niger.On the basis of acclimatization procedure and single factor, the optimal fermentationcondition of producting phytosterol was obtained by central composite rotate design. Onthis basis, through the comparative analysis five different extraction method incomeplant sterol content, and selected the best extraction method.The main results are asfollows:1. The Liebermann-Burchard colorimetric method and high performance liquidchromatography (HPLC) method were quantified the content of phytosterol; Throughimproved the Liebermann-Burchard colorimetric method: the time of chromogenicreaction was adjusted to24mins and the wavelength for recorded absorb value waschanged to610nm. Through establishment the HPLC analysis method: the UltimateXB-using C18column chromatography, mobile phase choose methanol:acetonitrile=35:65, wavelength200nm, column temperature:28℃, flow1.0mL/min.2. Through designed8groups different decay condition of bamboo shoots residue,chose the best source strains to continuous multistep culture. Using Specific culturemedium to isolated strains, and got9kinds of strains. The most powerful phytosterolstrains was selected, and identification was Aspergillus niger.3. Optimized the fermentation process. The results of the study show: under theconditions of acclimatization procedure3times, fermentation by Solid good raised,inoculation amount4.2mL/100g, fermentation time5.3d, fermentation temperature 33.3℃, nitrogen sourcechoose carbamide, add4.1%, the fermentation effect was thebest, and the content of total sterols was1168+32.4mg/100g in this condidtions.4. Five extraction methods were chose to extraction phytosterol from production,included Soxhlet extraction, ultrasonic cell disruptor extraction, refluence extraction,ultrasound aided extraction and traditional solvent extraction method. Throughdeterminated the total sterol content and compares their extraction yield, the resultsshow that the Soxhlet extraction>ultrasonic cell disruptor extraction>refluenceextraction>ultrasound aided extraction>traditional solvent extraction method, andcombining the actual extraction process for the energy consumption of the size,environmental problems, easy operation and other factors, choice ultrasonic celldisruptor extraction as the best method.5. The bamboo shoots residue as raw materials, total sterol and beta sitosterolcontent is complete index, then optimization the ultrasonic cell disruptor extractionmethod. The results of the study show that, ratio of material to liquid was1:31,ultrasound power was410w, ultrasound time was10.5min, raw material granularitywas86meshes, and under these conditions the total phytosterol extraction yieldwas85.6%, the beta sitosterol extraction yield was86.8%.
