This paper had a systematic research on the effects of Vibrio harveyi and dopamine (DA) on hemocyanins anabolism and phenoloxidase activity of Litopenaeus vannamei. Discussed mainly the following contents: (1) Effects of p-APMSF on the prophenoloxidase (proPO) activation system and phenoloxidase activity of hemocyanin of White Shrimp; (2) The combined influence of LPS,dopamine and p-APMSF on the prophenoloxidase activating system and hemocyanin-derived phenoloxidase activity of White Shrimp;(3) Effects of injecting Vibrio harveyi on the anabolism and hemocyanin-derived phenoloxidase activity of Litopenaeus vannamei; (4) Effects of dopamine injection on the anabolism and hemocyanin-derived phenoloxidase activity (HdPO) of White Shrimp.1. Effects of p-APMSF on the prophenoloxidase activating system and phenoloxidase activity of hemocyanin of White Shrimp. The paper reported the efforts of p-APMSF inhibitor on the phenoloxidase activity of hemocyanin of Litopenaeus vannamei haemocytes and hemolymph,through different concentrations of p - APMSF respectively with haemocytes and blood lymphatic. In the present paper ,we can see that p-APMSF is a effective phenoloxidase inhibitor but it has no effect on phenoloxidase activity of hemocyanin. And, through different concentrations of Settings, determine the p - APMSF optimal inhibiting concentration of 10μmol/L.2. The effect of combination of LPS,dopamine and p-APMSF inhibitors on the prophenoloxidase activating system and phenoloxidase activity of hemocyanin of Litopenaeus vannamei In this paper, we studied the effects of combination of LPS,dopamine and p-APMSF inhibitors on the prophenoloxidase activating system and hemocyanin-derived phenoloxidase activity of Litopenaeus vannamei. It was founded that they had significant effects on the PO, proPO and HdPO in White Shrimp (p<0.05), while the control had no marked difference. The hemolymph treated with LPS with p-APMSF and dopamine with p-APMSF in vitro showed conspicuous dose-dependent decrease in cell count. Phagocytic activity of haemocytes was promoted by LPS and inhabited by dopamine.3. Effects of injecting Vibrio harveyi on hemocyanins anabolism and hemocyanin-derived phenoloxidase activity of Litopenaeus vannamei In this study we founded that the effects of Vibrio harveyi and p-APMSF injection had notable effects on the hemocyanins anabolism, which included hemocyanin concentration, the expression of mRNA,p75- and p77-subunit,and PO activity of hemolymph, haemocyte and HdPO in Litopenaeus vannamei (p<0.05). The hemocyanin concentration and the expression of p75 and p77 subunits in the p-APMSF injected group showed peak changes at first. The result showed that p75- and p77-subunit and mRNA expression showed obviously increasing trend and reached the maximum at 24h, then they declined ,when at 48h they are still much higher than the control level (p<0.05) ,and the group that injected p-APMSF with Vibrio harveyi has the most obvious change. The result suggest that phenoloxidase activity of hemocyanin of every group no including the saline group showed obviously increasing trend and reached ,and the one which was treaded with p-APMSF and Vibrio harveyi still had much higher than others (p<0.05).The study showed that the hemocyanin concentration of p-APMSF injection group was lower than the control level but it has higher mRNA expression. So, it could demonstrat that under the invasion of pathogen White Shrimp started the hemocyanin synthesis and mRNA expression upward, and these hemocyanins mainly played immune functions, such as phenoloxidase activity, with other immune factors involved in immune response.4.Effects of dopamine injection on hemocyanins anabolism and hemocyanin-derived phenoloxidase activity of White ShrimThe study showed that the group which injected with p-APMSF and dopamine had notable effects on the hemocyanins anabolism, which included hemocyanin concentration, the expression of mRNA,p75- and p77-subunit,and PO activity of hemolymph, haemocyte and HdPO in Litopenaeus vannamei (p<0.05). Compared to the study about Vibrio harveyi injection we can see that there is almost the same trend between the two experiments. So it is suggested that there is some connection between the two. The result showed that the expression of hemocyanin mRNA still higher than the control level, so it maybe demonstrated that new synthesized hemocyanin mainly played immune functions, such as phenoloxidase activity, with other immune factors involved in immune response.
