Yet such as wheat scab and other wheat diseases seriously affect wheat yield and quality, which take the agricultural production and people's lives enormous damage. DON toxins from scab also endangered human food and animal feed safety and endanger the people and animal health hazard.gene tri101 is a major function genes who can resolve Fusarium graminearum toxin,it can not only reduce the incidence of scab,but also can reduce the mycotoxin accumulation to lower the harm of whert scab in the wheat yield and quality.so efficiently expressing of gene tri101 in wheat seeds can improve wheat resistance function and Ease harm of wheat scab toxins in growth,hoping Increasing wheat yield and quality, Producing wheat varieties with higher yields. This study fused gene tri101 and carrier pCAMBIA1304 and produced a carrier pCAMBIA1304-tri101 to transform wheat.we hope its efficiently expressing in wheat seeds improving wheat resistance function, easing harm of wheat scab toxins in growth, increasing wheat yield and quality and producing wheat varieties with higher yields. We selected a relatively new plants genetically modified method-Ion beam auxiliary method.and also discussed the influence of wheat emergence rate and the seed vigor by Vacuum processing and ion dose,so that we can explore a more perfect, swift genetically modified method to help transforming plant genetic and improving new varieties. The main results of this study are as follows:1.The building of the wheat seeds transformation expression vectorfusing gene tri101 and carrier pCAMBIA1304 and producing a carrier pCAMBIA1 304-tri101. shifting the to Plant expressing carrier in Agribacterium tumefaciens LBA4404 by freeze-thaw method2.The research of the influence of different ion implantation parameters on wheat emergencing Through discussing the influence of wheat emergence rate and the seed vigor by Vacuum processing and ion dose,we found that the influence of Vacuum treatment on wheat emergence rate and the seed vigor is not too big but the influence of Ion dose on wheat seedling emergence is obvious.Through comparing,we got the optimal parameters to shifting gene in wheatvacuum 10-2~10-3pa,energy 30 kev,beam 200mA,pulse width 400μs, frequency 25Hz,dose 8×1017N+/cm2, vacuum processing time 2~3h.3.
