Selenium (Se) is an important, biologically necessary trace element, which plays important biological roles in avian growth, reproduction, immune function and disease resistance. Se deficiency can to be correlated with the incidence of Keshan desease and Kaschin-Beck disease in human and white muscle disease of livertock. Selenoprotein is the main form of Se. Se exerts its biological effects mainly through its incorporation into selenoprotein. Levels of Se intake are associated with the change of selenoproteins and involved in the regulation of selenoprotein. Selenoprotein (W) SelWis an important member of selenoprotein family. The first observed function is that selenoprotein is associated with the mechanism of animal Se deficient disease, which receives extensively attention. At present, the studies about SelW are mainly carried out in mammalian, relatively little is known about the effect of dietary Se on the expression SelW in the livers of avian species.In this study, embryos chicken and cock were used in the experiment. (1). 0d embryos were sepreted to 3 groups, content of selenium in Control group is 0.07μg/ml, Se-I group is 0.08μg/ml and Se-II group is 0.10μg/ml, the livers of embryos were collected at 12,15,18,21 days. (2). One day old chicks were divided into 3 groups randomly, L group, M group and H group were fed dietary of Se as 0.033 mg/kg, 0.15 mg/kg and 1.5 mg/kg, liver tissues were collected at 15, 25, 35, 45 and 55 days. (3). One day old cocks were divided into 5 groups, control group was fed basal diet, Se-I, Se-II, Se-III, Se-IV group were added Na2SeO3 to the diet, the final concentrations of selenium were 0.6 mg/kg, 1.1 mg/kg, 1.5 mg/kg and 2.0 mg/kg. liver tissues were collected at 90 days.Real-time polymerase chain reaction was performed to detect the effect of Se content to the expression of SelW, SecS, SPS-1 mRNA. In addition, apoptosis gene Caspase-3, Caspase-8 and Fas mRNA expression were detected.,the results was displayed as follows:1. By the observation of microstructure of livers, it proved that selenium deficiency could cause the livers damage, and display on disorder of hepatic cord, inflammatory cell infiltrate and necrosis.2. By the detection of the content of Se and SelW,SecS,SPS-1 mRNA expression levels in chicken livers, it discovered that SelW,SecS,SPS-1 mRNA ecpression levels were regulated by the content of dietary Se. It indicated that Se concentration in the dietary could interfere in the gene transcription of tissue SelW,SecS,SPS-1 mRNA.3. By the detection of the liver cells apoptosis and expression level of Caspase-3, Caspase-8, and Fas mRNA in livers, it discovered that selemium deficiency could induce the liver cells apoptosis, increase the expression level of these genes. It indicated that apoptosis probably played an important role in the development of liver damage induced by selenium deficiency, which was induced by the cascade reactions of Fas/FasL-mediated Caspase family...
