The research object in this paper is Pseudosciaena crocea. First, In the condition of MS-222 anesthesia, the study is about the keeping alive technology of the pseudosciaena crocea and its influence on Physiochemical indices and haematological indices; then, we formulated kides of Freezing point regulators to provide a preservative for lowing the Freezing point of Pseudosciaena crocea.Fitst, The anesthesia technology transportation of Pseudosciaena crocea was examined under laboratory conditions. on behaviors of Pseudosciaena crocea were examined at the temperature of 10~11℃, including changes of characteristic behavior during anesthesia and recovery process,breath frequency and the effect on anesthesia and recovery times and the role of temperture on anaesthesia time. Meanwhile, considerd as factors of Anesthesia concentration, Transport temperature and water ratio, the formulation was optimized using orthogonal array design. It showed that the effective concentration was 50~60 mg/L and the higher the concentration was, the bigger the drop rate of the breath frequency was. Meanwhile, When the temperature reached 16℃, the anesthesia time was most stable and the Tlm of 30 min was 55 mg/L respectively. By the way, 11-13℃is the best transport temperture and its Limit death temperature is 7℃. The small the ratio was, the long the keeping alive time, however, when the sea of added quantity achieved certain limit, keeping alive time had little effect. The last, the optimal process of keeping alive technology was determined that under 16℃, MS-222, which concertration was 45 mg/L ,as an anesthetic to soaked Pseudosciaena crocea in 5 min, then the fish were put in 11℃sea water as ratio was 1:3, at the same time, we must make sure the transport temperture is 11℃.Then, The physiological indexes and biochemical indexes of were determined and analyzed statistically during all anesthesia and keeping alive process. Changes of theblood indexes of Pseudosciaena crocea are as following: erythrocyte count and packed cell volume improved with significant difference (P < 0.05), and significant difference was found on parameters of hepatic glycogen, lactic acid and creatinine(P <0.05. On adverse, the pH, Fat content, total protein, albumin, alanine aminotransferase, aspartate aminotransferase, lactate dehydrogenase, blood glucose and total cholesterol did't change significantly. Which means keeping alive technology of the pseudosciaena crocea with MS-222 anesthesia would not destruction nutritional value and commercial value, and the damage ro liver cells and myocardial function is lesser, but it had partial suppresses kidney function.Last, a preliminary study through used of different kinds of freezing point regulators and complex to lower the freezing point of pseudosciaena crocea. The formulation was optimized using Box-Behnken response surface analysis design. As a result, A blend containing 3.80 % CaCl2, 2.09 % NaCl and 4.08 % provided an optimal inhibitory effect against deteriorating of Sensory quality , growing of total bacterial count (TBC) and increasing of total volatile base nitrogen (TVB-N) were periodically measured during superchilled storage.
