Forest ecosystems is the main terrestrial ecosystems which is widely distributed and has many functions. The natural forest succession or disturbance activities have the characteristics of spatial distribution which makes the distribution of trees with a high degree of spatial and temporal heterogeneity. The classical statistics mainly studies "Properties" feature, generally not to involve the spatial structure of "properties". Spatial statistics which keeps a close connection with geo-statistics having originated from geology is always widely used and based on classical statistics. Spatial statistics is based on spatial autocorrelation of investigated objectives and connected with the spatial variability of investigated objectives. It can reduce costs and improve efficiency to apply spatial sampling techniques.In this paper, the sample survey objective is the area of forest cover in Huaxi district in Guiyang City. The research selects SPOT5 images as the original fusion image. Sampling frame layout is based on the fusion image. In the same conditions, to use a simple random sampling, a systematic sampling, a spatial random sampling and spatial stratified sampling and to compare the sample results can be concluded:1) Based on sampling theory, spatial stratified sampling survey of forest resources is the best sampling method. Spatial data with spatial coordinates almost have a certain spatial correlation and forest resource data as well. Spatial sampling recognize the spatial correlation and to quantify this correlation. In the sample it makes decision of sample by this correlation to reduce sampling costs.2) Based on the same level of prior sampling, making the sample size as the sample cost index, the cost of spatial stratified sampling is the lowest, followed by spatial random sampling, and finally the classic sample. It creates a certain degree of sample redundancy that classical sample don't consider spatial autocorrelation of survey objectives.3) The survey gave a spatial sandwich sampling on the basis of spatial stratified sampling. Spatial sandwich sampling is the reverse operation of spatial stratified sampling. Based on not expanding the sample size, it may generate a report in any place of interest to investigators in the survey zone.With the continuous development of 3S technology and the maturity of spatial statistics, spatial sampling techniques will be used more and more for the sampling survey of forest resources.
