| Tobacco is one of the most important cash crops in the world, and China occupies the first planting area of tobacco in the world. It is reported that, the main virus disease, which includes Tobacco Etch Virus (TEV), Potato Virus Y (PVY), Tobacco Mosaic Virus (TMV), and Cucumber Mosaic Virus (CMV) and so on, has caused large losses about tobacco production and has become an urgent problem to be solved. Therefore cultivating and using the high quality disease-resistant germplasm resources is the most effective prevention measure and economic mean in the field of controlling tobacco virus disease, and it has a very important practical significance.This study obtained the pure TEV and PVY, and identification of resistance against TEV and PVY was carried out for 21 tobacco cultivars by using artificial mechanical inoculation. Seeding is in mid-march of every year, vaccination is in Late June, the inoculum is 3g diseased leaves plus 100ml0.01mol/L phosphate buffer (pH = 7.0). Tobaccos were inoculated with virus, when they showed symptoms and began the general investigation, then calculating disease resistance was performed based on investigation data. The results are as follows:Among these materials, disease resistance to TEV, Shuangkang70 and Zhongyan103 were resistant; Yunyan97, Qinyan96, Liaoyan17, Yunyan203, K326, Qinyan98, Yunyan85, G28, Zhongyan90 and Longjiang237 were moderate resistance; Other 9 sensitive were NC89, Hongda, G80, Yunyan87, RG11, jinyehuang, CV87, Coker176 and jinxing6007.Among these materials, disease resistance to PVY, Liaoyan17 and jinxing6007 were resistant; Qinyan98 and Shuangkang70 were moderate resistance; Other 17 sensitive were NC89, Qinyan96, Yunyan87, jinyehuang, G28, Yunyan85, Hongda, G80, K326, Coker176, Longjiang237, Yunyan97, RG11, Zhongyan90, Zhongyan103, CV87 and Yunyan203.In this study, the results identified can not only be utilized to provide the basis for selection and variety deployment, but also as parent materials in breeding for resistance and lay the foundation of prevention research. |
