This article do transgenic research on Torenia fournieri Lind. with HearNPV Chitinase and maize Lc transcription factor genes, mainly discussing the expression of exogenous genes. The study is to improve the contents of Chitinase and flavonoids by means of transgenic technology. And lay the Foundation for other transgenic research of plants. Major findings are as follows:Through the method of PCR amplification, HearNPV Chitinase and maize Lc transc- ription factor genes have been obtained and cloned into plant expression vector pBI121.The recombinant plant expression vector ChiA-pBI121and Lc-ChiA-pBI121have been constructed and turned it into Agrobacterium LBA4404 successfully. By Agrobacterium-mediated genetic transformation of torenia leaf explants, 8 geneticallymodified strains have been obtained, named as C1, C2, C3, G2, G3, A1, A2, A3. Semi- quantitative RT-PCR analysis showed that the ChiA gene mRNA expressions of 8 genetically modified strains are significantly higher than the control group (P<0.05). Between these 8 strains, C3 has the highest mRNA expression while A2 is the lowest one. It has been proved that HearNPV ChiA gene has expressed normally in torenia. The chitinase activity of C1, C2, A1 and A2 is higher than the control group and A1 has the highest activity. In these 8 genetically modified strains, the flowering of 7 strains is earlier than others.In addition, 3 resistance bud of transforming Lc and ChiA genes have been obtained after a long-term selection. But it is a long term to analysis their expression levels and interactions of those genes. The plant transgenic research with transforming two genes has laid a good foundation to select high performance disease-resistance plants.
