Duck viral enteritis (DVE) caused by DEV., also known as duck plague, is a febrile,acute,contagious disease of anseriformes waterfowls,such as ducks,geese, and swans DEV gives rise to the hemorrhagic button or band-like lesions found on the mucosal surface of the intestines,vascuLar damage, degenerative changes ,and caseous exudate in trachea and cloaca.DEV is easily able to establish latency in the trigeminal ganglia and lymphoid tissues except for acute infection.DEV is composed of a linear, double stranded DNA genome.The genomic organization is UL-IRS-US-TRS,and was found to be 158Kb in length,and encodes78 proteins.DEV which corresponds to Iltovirus and Varicellovirus, is classified to intermediate taxonomic entity in the subfamily Alphaherpesvirinae. Herpes virus with more nonessential genes can contain the big size of the inserted sequences,and is good live virus vector for expressing foreign gene. The recombinant duck enteritis virus with the GFP expression cassette inserted to the local of between UL45 and UL46 gene was vaccinated SPF duck and, the copies of rDEVuL4546gfp DNA in Brain, Thymus, Spleen, Kidney, Liver ,Blood and Bursa of fabricius of vaccinated SPF duck by rDEVuL4546 was the determine index which developed the research on the law of dynamic distribution of rDEVuL4546 in SPF duck.The specific primers (RTF and RTR) were designed according to the UL6 gene of GenBank and the standard curve was established using SYBR Green I real-time fluorescent quantitative method. Copies of the virus DNA were quantified in collected tissues by the standard curve,the rDEVuL4546 and DEV DNA copy number were carried on the analysis of significant difference using SPASS 16.0 software.The resuLts of this investigation displayed that the standard real-time fluorescent quantitative method detection of DEV was established.DEV and rDEVuL4546 with pan-tropic mainly distributed in Brain,Liver and Kideny. The GFP expression cassette was inserted to the local of between UL45 and UL46 gene.That is no effect on reproduction and distribution of DEV C-KCE in SPF duck, except for the copies of rDEVuL4546 and DEV DNA were significant difference in specific times and there was no significant effect on reproduction of DEV inserted foreign gene in immunized SPF duck. It indicates that duck enteritis virus is to be a live virus expression vector for foreign gene.
