The Genetic Polymorphisms Of Chicken MHC Class Ⅰ Genes

Chicken MHC class I genes are highly related to genetic resistance to many kinds ofinfectious diseases. To provide molecular genetic basis for chicken breeding, furtherunderstanding of the chicken MHC class I molecules encoded by BF1and BF2genes is aperquisite. All56samples of experimental chicken lines carrying known MHC haplotypesand indigenous chicken populations having unknown genetic backgrounds of MHC BF geneswere included for a depth investigation of the complete BF gene genomic DNA structures.The major results are as follows:(1) BF1gene deletion phenomenon was proved to be true inchicken in two different types, one to be a deletion heterozygous of BF1gene and the other adeletion homozygote of BF1genes, which length was3196bp from the first base of EnhancerA to-187bp upstream of TAP1gene;(2) BF1gene was divided into three categories, such asA, Adeland B, there were six genotypes of AdelAdel, AA, AAdel, AB, AdelB and BB,respectively;(3) It established two methods to amplify the BF1genes independently forscreening the deletions of BF1genes, and one method for sequencing BF genes simply;(4) Itwas impossible to specifically amplify the exons2or3of BF genes before screening thedeletions of BF1genes, and also cannothe't divide into many fragments to research, thelength of PCR products should be longer than3511bp;(5) Totally, it got overall length ofBF1and BF2genes, obtained22new haplotypes by nominating two genes together, of which15haplotypes were shared by BF1and BF2genes, four were unique BF1haplotypes and theremaining three were unique BF2haplotypes;(6) The overall length of BF1gene wasenlarged from2052bp to2058bp, and variable sites enlarged from186to236, of which50mutations were29in CDS, the other21were in NCS;(7) The overall length of BF2genewas enlarged from2026bp to2035bp, there were also236variable sites complete BF2geneexcluding intron3, of which variable sites47were enlarged in CDS.
