The experiment uses Vitis piasezkii Maxim. for test materials,with plant tissue culture technology of growth medium for its research; And with frost-resistant different varietals tube seedlings leaves as research object,the will of the grape hardiness different 4 respectively tube seedlings,25℃,4℃,0℃and -4℃,under the condition of treatment 4 hours cryogenic stress were determined,and then leaves of chlorophyll,soluble protein,soluble sugar,of mda and free proline,peroxidase,catalase,superoxide dismutase (SOD) content changes in activity and root vigor; Finally,the application of grafting in plant Vitis piasezkii Maxim .micro tube seedlings leaves grapes for stocks,with other frost-resistant different varietals tube seedlings for different varieties of the scion test-tube grafting,which aims at revealing frost-resistant different varietals tube seedlings physiological and biochemical characteristics of the differences,and screening get frost-resistant better grape varieties and micro grafting combination,for grape hardiness research and provide theoretical basis for cultivation. Research shows that:1.The optimum medium for Vitis piasezkii Maxim initial culture was GS+KT 0.5 mg/L+6-BA 0.5 mg/L+NAA 0.5 mg/L,the optimum culture medium for subculture was GS+NAA 0.1mg?L,the rooting medium was GS+IBA 0.2mg?L,the most optimum transplant strom were fertile soil and saw dust and organic fertilizer .2.Compared the survival rates among the four combination(Red Globe/ Vitis piasezkii Maxim,Phexi/ Vitis piasezkii Maxim,Cabernet Sauvignon/ Vitis piasezkii Maxim,Merlot Noir/ Vitis piasezkii Maxim),the survivals of the combination(Red Globe/ Vitis piasezkii Maxim)were better than Phexi/ Vitis piasezkii Maxim and Cabernet Sauvignon/ Vitis piasezkii Maxim,the poorest one is the combination of Merlot Noir/ Vitis piasezkii Maxim.3.Grape tube seedlings after treatment by low temperature,especially -4℃low-temperature tube seedlings after treatment,grape leaf chilling injury symptoms. The chilling red globe grape tube seedlings its direct performance:tube seedlings leaves,leaf appeared a stain shape spots,leaf retreat green turn yellow and happen Browning,especially leaf edges symptoms more evident;Leaf break the skin flow glue,fall off,falls,2 bits of face first appeared in young leaves,macular W.P.A pointed inward curly,wilting. The variable leaves chilling LDP294 tube seedlings relative red grape vine is concerned,its blade earth W.P.A shape phenomenon is not obvious,the blade has not occurred back green,a slight Browning;Young leaf blade inward curly,wilting. 4. After treatment,whether by low temperature of hardiness strong or weak grape variety tube seedlings,MDA content in 25℃~ 4℃,0℃~ -4℃increased with the temperature reducing,at 4℃~ 0℃scope with temperature decrease,free of proline,soluble sugar content increases linearly with temperature drop,soluble protein in 25℃~ 4℃scope with temperature decrease,chlorophyll content and root activity decreased,relative electric conductivity increases,POD,the activity of SOD and CAT was significantly improve,when the grape varieties tube seedlings in continuous after four℃and 0℃low-temperature processing,its leaves CAT and SOD activity within the POD activity,but obviously improve the improve only in tube seedlings after 4℃processed;By 0℃in processing temperature to reduce - 4℃in the process,grape tube seedlings of the activity of SOD and CAT with a certain degree of POD activity decreases,and it has improved. Different varietals tube seedlings after low temperature treatment within the organization cells after the protective enzymes improve degree is also different. In the low temperature treatment process,the cold hardiness of hair becomes less strong within LDP294 blade vine leaves free proline,soluble sugar,soluble protein content,chlorophyll content,root vigor,relative conductivity were higher,and hardiness sent red earth of physiological and biochemical indexes of leaves are low,so content of the LDP294 variable leaf grape hardiness than red earth. The measurement of the different grape varieties in tube seedlings with their metabolic substances content changes the consistency of the cold hardiness,can explain the metabolic substances as early with plant hardiness indexes of judge plant hardiness is feasible.
