Haploid is good material of somatic cell genetic research and mutation breeding.through chromosome to redouble in haploid plantlet,it can obtain double haploid,which is Nondisju -nction,DH population was good material for the molecular of AFLP,RELP,RAPD,SSR marker and genetic map construction.this reaserch do some work on in vitro gynogenesis to induce haploid plants in cucumber, and construction of genetic map with SSR markers, the construction population was DH population come from irradiated pollen induce techno -logy. the results are as follows:1.Effect of genotypes on haploid plant induced by pollination with irradiated pollens was studied, by using 29 cucumber genotypes as female. the results indicated that variation of induction efficiency ranged from 0.18 to 2.20 and the mean was 0.76. 11 genotypes haploid plan induction efficiency ranged from 0.5 to 1,which occupied 37.93% among all genotypes.10 genotypes haploid plan induction efficiency ranged from 0.18 to 0.5,which occupied 34.48% among all genotypes. induction efficiency of different genetypes had diversity.2.Effect of genotype,TDZ and AgNO3 on haploid plant induced by gynogenesis was studied.the results showed that genogensis initiaton frequency of different genetypes had diversity. 9 cucumber genotypes variation of induction frequency ranged from 65.52% to 93.33%;initiation of genogensis the optimal TDZ is 0.08mg/ L in inducting medium;Adding AgNO3 to inducting medium could increase Ovule initiation frequency,induction optimal AgNO3 concentration is 5mg/L;result 8 plants were regenerated in this research,8 regenerated plant are haploid,2 regenerated plant are diploid through analysis of Flow Cytometry.3 with 75 DH lines as population,and by female parent P281 and male parent P282 were used to construct the cucumber genetic map.the map consisted of 4 linkage groups,it spanned a total genetic distance of 187.963cM, with an average map interval of 17.35cM between two markers. the maximum in average distance between markers 14.914cM,the length of linkage group is 17.038-71.605cM,avery linkage group has 3-29 loci,and average genetic distance is 6.39-31.99cM.61.19% of the markers exhibited distorted segregation and significant level or extremely significant level .
