The molecular technique of random amplified polymorphic DNA (RAPD) was used toinvestigate the genetic diversity of4Phyllostachys edulis populations from Jiangsu and Zhejiangprovince. The genetic diversity and genetic relationships, and differentiation of these populationswere evaluated and discussed with several analysis methods of modern population genetics. Thespatial distribution of Phyllostachys edulis population is also be investigated by the method ofspatial molecular ecology. The purpose is to understand the level of fine-scale genetic diversity,the source of differentiation, spatial distribution of cloning and population history dynamic onPhyllostachys edulis populations. Moso bamboo′s sampling strategy is also preliminarily studied,which further prove the relation between breeding system and genetic structure and provide atheoretical basis for studying the adaptive evolution about bamboos. What′s more, it providesbasic information of relations between environmental factors and genetic variation or clonalreproduction. The main results are as follows:1. The result of RAPD indicates that the level of fine scale genetic diverstiy within4Phyllostachys edulis populations is low. Nei′s gene diversity (Hs) is0.0048, Shannon′sinformation index (I) is0.0075while the percentage of polymorphic bands (PPB) is2.1%. Butthe value is higher at population level. Three indexes of genetic diversity are as follows: Ht=0.0180, I=0.0279, PPB=8.4%.2. Gene differentiation coefficient (Gst) among populations is0.7333, gene flow (Nm) is0.1810. It shows that genetic variation mainly exists in different populations.3. Phyllostachys edulis has a middle level of clonal diversity. The mean value of Nc, G/Nand D are11.5,0.54and0.14respectively. Four investigated populations are all multi-clones and10genotypes are local while no widespread genotypes were found. It indicates a obvious clonaldifferentiation between fore populations.4. According to the results of genetic diversity analysis, we guess that the4genetypes ofNY1are evolved from G1by somatic mutation; bamboos of NY2population may betransplanted from TH; and there was probably an population regeneration occurrenced at AJpopulation.5. The clonal architecture of Phyllostachys edulis is guerilla and the maximum distance agenet can reach is1448m.6. When sample size reaches50-60, the sample population includes more than90%geneticvariation of a population. So60is the adaptive sample size for Phyllostachys edulis population.7.65m is a suggest distance which should be kept as intervals among individuals forPhyllostachys edulis sampling.
