| Lily varieties bred has considerable ornamental value and commercial value,but it wastaken times from hybridization to process the seed-bulbs to beginning flowering, at least3to5years, and even more than ten years to be fostered. Apparently,it is imperative to develop anearly and effective identification method for authenticity of lily hybrids. In this research, themolecular cytogenetics methods, as Gimesa C-banding, fluorescence in situ hybridization with45S rDNA probe and genome in situ hybridization, were applied to identify hybrids. The mainresults were follows:1. LA hybrid 'Royal Lace' and Asiatic Lily 'Golden Horn','Pollyanna','Brunello' were hybridparent materials. They were four combinations to be carried out,'Golden Horn'×'Pollyanna'(2x×2x),'Golden Horn'×'Brunello'(2x×4x),'Royal Lace'×'Pollyanna'(3x×2x),'Royal Lace'×'Brunello'(3x×4x), and different numbers of offspring from the combinations were obtainedthrough embryo rescue.2. Four aneuploid and three pentaploid hybrid progenies from 'Royal Lace' x 'Brunello' andthe parents were to be checked. The C-band existed on most of the chromosomes of parents, andpolymorphisms of the C-band pattern were shown between the homologous chromosomes. Therewere11and18signal sites of45S rDNA FISH on 'Royal Lace' and 'Brunello' respectively, andwere from14to21signal sites in offspring. Typical chromosomes with parental signal sites canbe observed among progenies. It was confirmed that the offspring measured are true hybrids.3. Distant hybridizations have been conducted between Oriental Lily 'Constansa' and L.regale. Three diploids progenies were to be analyzed by the early identification techniques. TheC-banding patterns appeared on chromosome among offspring were quite similar related to thebanding patterns of their parents. And all signal sites combined with45S rDNA fluorescence insitu hybridization near the centromere (11for 'Constansa' and12for L. regale), can be observedin the offspring, either11or12. Each offspring contains labeled chromosomes from their parents.Genome in situ hybridization results were even more intuitive and clearly distinguish the parentalgenome in the progeny.4. For the hybridization between the different ploidy lily varieties, hybrids can be initial identifiedby the phases of cell mitotic chromosome, in which the chromosome numbers shown the differencesbetween offspring and parents. For distant hybrids, genomic in situ hybridization may intuitiveaccurately distinguish the parental genome, more efficient and effective. |
