β-defensin has a broad-spectrum microbicidal activity against mastitis and was also an important participant of Innate immunity. So the research ofβ-defensin is very useful to prevent against and cure mastitis. In this experiment, we explored changes of Defb1 expression during developmental cycle(pregnancy, lactation, nature delactation) of mouse mammary gland, and explored the effect of mastists which caused by LPS to the expression of Defb1 in mice mammary gland by semi-quantitative method.ⅠThe levels of Defb1 mRNA in mammary gland of mice. A pair of cloning primers were designed. Total RNA extracted from mice mammary gland was amplified by RT-PCR, the PCR products were ligated into the Pmd19-T vector, then transformed into E.coli DH5αcompetent cells. The positive clone was identified and the sequence was sequenced, analyzed and structure prediction. The results show that Defb1 gene can be expressed in mammary gland of mice.ⅡDefb1 expression changes during developmental cycle of mice mammary gland: We got the fourth breast tissue of mice at the 1d, 6d, 12d, 18d of gestation; 1d, 6d, 12d, 18d of lactational and 0d, 1d, 2d, 3d, 4d, 5d after natural weaning. Changes of Defb1 expression were studied by semi-quantitative. The results show that: the expression ofβ-defensin1 is gradually increasing with the development of pregnancy and the difference is remarkable; With the process of lactation, Defb1 is gradually decreasing. The difference between Group 1d and 2d is not significant. Other groups have significant change compared with lactation 1d. Compared with 1d at natural delectation, the expression ofβ-defensin1 is significantly decreased at 1d, 2d and increases sharply at 3d, but decreases at 5d.ⅢThe expression of Defb1 in mammary gland which were treated by LPS: LPS were injected into the base of the fourth mammary gland of mice which were 10d after parturition. We got the fourth breast tissue of mice at the 0h, 6h, 9h, 12h, 24h after injection and analyzed by semi-quantitative. The results show that: the expression ofβ-defensin1 is significantly increased at 6h and 9h Compared with 0h and the max expression is appeared at 9h. Then expression is significantly decreased at 12h and 24h, but compared with 0h, the expression is significantly increased at 12h and 24h. Key words: mice; mammary gland; Defb1; LPS...
