Creatine pyruvate (CrPyr) is a novel multifunction nutrient supplements, with both the function of Pyruvate and the characteristic of Creatine. It's main compenents is Pyruvate and Creatine.The main functions of CrPyr are to promote energy production and endurance to maintain, improve exercise capacity, rebuild energy reserves to maintain health, shorten the recovery time after fatigue and increase muscle weight. At present, most research has been focused on Sports Physiology. The precise mechanisms by how CrPyr exerts its biological actions in amino-acids metabolism are not fully understood. Therefore, a study was carried out to examine the effects of exogenous added CrPyr on amino-acids metabolism in adult male AA broiler chickens. The purpose of this study is to provide a theoretical and experimental evidence for CrPyr regulation on amino-acids deposition in AA broiler chickens.1 The method of Determination of Amino-acids by Reverser-phase High Performance Liquid ChromatographyA pre-column deviation procedure of O-Phthaldialdelhyde (OPA) was used for the determination of amino-acids. Amino-acid derivatives were separated on Waters Spherisorb ODS (4.6 mm×250 mm,5μm) column. Gradient elution was adopted with fluorescence detection. Fifteen amino-acids were separated and detected within 35min. Linear relationship between the peak area and concentration in the range of 5.20 to 333nmol·L-of the amino-acids, with correlation coefficients of 0.9991 to 1 was obtained. Amino-acids with the intra-day and inter-day RSD are 2.32%-5.02% and 4.20%-5.90%, respectively. The lowest detect area is 1.2nmol·L-1 The results show that there are good linearity and regression,and the method can be used to determine the nature and fix quantify.2 Effect of pyruvate, creatine and creatine-pyuvate on profile of amino-acids in partial tissues of broiler chickensIn this study,four hundred AA broiler chickens born after 21 days was divided 4 groups randomly, four replication each group, and 25 AA broiler chickens each replication.The control group (C) was gave common animal feeds, and the trait groups were to supplement 2% pyruvate (T-I),3% creatine(T-Ⅱ),5% creatine-pyruvate(T-Ⅲ) in common animal feed. Fed 42 days,4 broiler chickens were killed randomly in each replication, collecting blood and separating serum, getting liver and leg muscle. Then we detected amino acids in serum,liver and leg muscle by RP-HPLC. Fifteen amino-acids were separated and detected within 35min. The results show that pyruvate, creatine and creatine-pyruvate can significantly decrease Thr and Tip in serum, pyruvate, creatine and creatine-pyruvate can significantly increase Arg and Val in leg muscle, and pyruvate, creatine and creatine-pyruvate can significantly increase Asn, Thr, Tyr and Val in liver. This study conclude that pyruvate, creatine and creatine-pyruvate can indirectly change the direction of Nutritional material in AA broiler chickens, with accommodating amino-acids content.3 Effect of pyruvate, creatine and creatine-pyuvate on relative enzyme activity in broiler chickensThe trial design and sample collection are same as the chapter 4. The activity of GSH-PX. CAT, T-AOC, MDA in serum and liver were detected by kit, GOT and GPT in liver and CK in leg muscle was detected,also. The result show that creatine-pyruvate can decrease better activity of GSH-PX than creatine; All of pyruvate,creatine and creatine-pyruvate have T-AOC ability, but creatine-pyruvate have higher activity than pyruvate and creatine. creatine-pyruvate elevate ability of CK better than creatine,and creatine elevate ability of CK better than pyruvate. This study conclude that the function of anti-oxidant,regulating amino-acids metabolism and energy metabolism of creatine-pyruvate is better than pyruvate and creatine in AA broiler chickens.
