Xanthomonas oryzae pv. oryzae (hereafter Xoo) is one kind of plant pathogenic bacterium that causes bacterial blight disease in rice. Bacterial blight is a very serious disease of rice.Xoo China 13751 strain XOO2193 genes DNA sequence completely consistent with Xoo MAFF311018, Xoo PXO99A, Xoo KACC10331 the genome that encoding 6-Phosphogluconolactonasgene。6-phosphogluconolacton ase is the important components in HMP pathway,catalyzes the second step reaction of HMP pathway,catalyzes 6-phosphogluconolactone to 6-phosphogluconate, this reactian is Hydrolysis. HMP pathway is one of saccharide decomposition metabolic pathways which is widespread in animals, plants and microorganism.The main characteristics of this pathway is that glucose can completely be oxidized not through the TCA cycle and EMP pathway and produces a large number of NADPH and a variety of important intermediate metabolites.In order to study the function of the XOO2193 gene, we constructed nonpolar mutant GNM2193 by the suicide plasmid pK.18mobGⅡ, and integration a complementary strain GNC2193. Phenotype analysis of the mutant GNM2193 that the virulence of mutant GNM2193 significantly reduced,the mutant loast the ability to cause hypersensitive response on the non-host plants (Ricinus communis), and decreased production of extracellular polysaccharide. Complementary strain GNC2193 that virulence, hypersensitive response, production of extracellular polysaccharide recover to Wild-type level. In addition, it is found that the growth of the mutant in rich medium OB and in minimal medium XOM2 was slow. Mutant growth slow on host plants rice.The mutant GNM2193 utilize carbon sources ability was decrease.All these findings indicate that XOO2193 gene plays an important role for the growth and the virulence of Xoo.
