The chemical composition and biological activity of the protein bait of Primabait which was made by Pupuk Alam Company of Malaysia and GF-120(alsonamed hunting flies0.02%bait agent) which was produced by Dow AgroSciencesand the protein baits for Bactrocera dorsalis (Hendel) which was produced by ourresearch institute was studied in this thesis. The active ingredient of the protein baitswas discussed. The results are as follows:1. The basic properties analysis of protein baitsThe pH and the content of solid, moisture absorption properties, viscosity, andcontents of heavy metal materials were compared within three proteins. The resultsshowed that: the pH of GF-120was3.85, its solids content was43%and its viscositywas5.721×10-3. The pH of Prima bait was3.67, its solids content was23%and itsviscosity was1.036×10-3. The pH of the protein baits for B. dorsalis which wasproduced by our research institute values of the largest was5.70,its solids content was33%and its viscosity was1.263×10-3. The bibulous rate of the three protein baitsdepends on the watercontent of the environment. The protein baits were morebibulous when the humidity was higher. The percentage of moisture absorbedincreased heavy with the rise of temperature when the humidity was under70%. Theabsorbing moisture percentage increase heavy with the rise of temperature rise againin down first when humidity above70%. The content of metal elements in GF-120was the highest, the protein baits for B. dorsalis which was produced by our researchinstitute content was the lowest, and the Prima bait in the middle.2. The composition analysis of protein baitThe content of protein in the three protein baits was tested by the Coomassiebrilliant blue method and kieldahl method, the former by drawing standard curve, thencalculated the content of protein. The content of protein for GF-120was25.5mg/mL,which was the highest. The content of protein for Prima was5.8mg/mLl which was the lowest. The content of protein for the protein baits for B. dorsalis which wasproduced by our research institute was6.6mg/mL,which was in the middle; However,when the kieldahl method was used to test the content of the protein, the proteincontent of GF-120was5.4mg/g, which was the lowest, the protein baits for B.dorsalis which was produced by our research institute for the highest protein9.4mg/g,and Prima is in the center for8.9mg/g. the Coomassie brilliant blue method uses astandard protein to determine the unknown proteins whereas the kieldahl methodmeasures the nitrogen content to determine the content of the protein. So, the kieldahlmethod is more accurate.The SDS electrophoresis was used to measure the protein molecular weight inthe protein bait, which shows the protein molecular weight in GF-120is above20KD.Most of the protein molecular weight in Prima bait and the protein baits for B.dorsalis which was produced by our research institute are less than26ku. The resultshowed that a protein strap can be observed at26KD position in Prima.High Performance Liquid Chromatography with Precolumn Derivatization wasused to analyse the kinds and contents of amino acids in three protein baits for B.dorsalis. The results showed that there were8kinds of free amino acids in GF-120and the other two protein baits contains17kinds of free amino acids and three proteinbaits contains18total amino acids.The total amino acid in GF-120was1762.34mg/100mL and free amino acids was255.68mg/100mL, peptides contents1506.66mg/100mL; Prima protein bait contains for5284.76mg/100mL total amino acid and573.64mg/100mL free amino acids, peptides contents4711.12mg/100mL; Theprotein baits for B. dorsalis which was produced by our research institutecontains630.10mg/100mL of free amino acid, total amino acids for6562.43mg/100mL, peptides contents5932.33mg/100mL.the content of the total reducing sugar in the protein bait was analyzed, theresults showed that GF-120contains the most, for28.2g/100g, the second is Primacontains9.1g/100g, The protein baits for B. dorsalis which was produced by ourresearch institute contains the smallest for only5.3g/100g.To increase the total free sugars of protein bait, Acid hydrolysis was used to get free sugars from protein bait.The influences of acid concentration, the reaction temperature and the reaction timewere analyzed using single factor experiment and orthogonal test. The results showedthat the optimum conditions were2.5mol/L acid at95°C for3h. The hydrochloricacid concentration was the most important factor influencing the hydrolysis processwhereas the reaction time showed the slightest influence towards the hydrolysisprocess.3. The biological test of the proteinFor three proteins, the attractive effect of Prima and the protein baits for B.dorsalis which was produced by our research institute were similar, and are muchbetter than GF-120.For different material of the protein bait the attractive effect of the three areequal. So the attractive effects of the protein bait are irrelevant to material.
