In this study, single spores of Volvariella volvacea were obtained by spore isolation from thestrain PYd15×PYd21, and then eight isolates with homokaryons were screened out for further researchby SV marker developed in this research. These eight single spores were cross with each other in thetest and identified by morphological marker and molecular marker. The result showed that the V.volvacea strain PYd15×PYd21was two polar mating type mushroom after mating type analysis.Eight isolates were divided into two groups according to difference of mating types termed A1and A2.Among eight isolates, V1, V4, V5and V17are in a group different from the other group consisting ofV2, V10, V14and V16.Mating tests between isolates V0115, V0116(female parent V0045); isolates V0121, V0123(female parent V0032); isolate D7(female parent V0158) and the standard strain (isolates with A1andA2factors which female parent PYd15×PYd21) were carried out and demonstrated that A factor typeof five isolates are different from that of standard strain, A≠A1and≠A2, according to analysis onmorphology and SV molecular marker. Together with four different types of mating type factor, Afactor types were identified among five isolates, there are6different types of mating type factor A,which suggested that factor A in V. volvacea shares the extensive polymorphism with other mushroom.We extend our knowledge on genome information of V. volvacea and PYd21-MIP sequence wassequenced, A mating site was identified nearby the MIP gene. Furthermore, A mating site wasconfirmed through protein sequence blast in NCBI. Based on PCR analysis amplifying mtA15andmtA21, isolates V1,V2,V4,V5,V10,V14,V16and V17from mating test bwtween PYd15×PYd21were divided into two categories, as A factor of V2,V10,V14and V16identical to that of PYd15, andfactor A of V1,V4,V5and V17identical to PYd21. However, the PCR analysis on determining Afactor of8isolates was consistent with mating tests, which further confirms the existence of matingtype factor A. The research also indicated that genes locating on both sides of mating factor A are quiteconservative. However, the sequence of mating factor A harbors extensive polymorphism that wasproved on genome wide level. The research on polymorphism of mating factor A in straw mushroomwill advance the works on breeding and genetic research.
