Microenvironment change of uterine local endocrine, is a dynamic process changedconstantly in response to oestrous cycle and pregnancy process. These processes are synergiceffects of steroid hormone, chemokine, cytokine, transcription factor, lipid medium, andplayed a unique role in endocrine and reproductive immune. As the main sex hormones ofuterine, estradiol(E2) and progesterone(P4) play a key role in prompting embryonicdevelopment and building the endometrial receptivity by regulating endometrial cells secreteactivity through their receptors. Therefore, studying the effects of E2and P4to endometrialcell secretory activity will provide new insights, about deep understanding fetal-maternalimmunoregulation, abnormal pregnancy and pathological uterine.In this study, based on the hanging cell coculture insert system, immortalization of goatsendometrial epithelial cells (EEC) and stromal cells (ESC), which obtained by transfection ofhuman telomerase reverse transcriptase (hTERT), has trained coculture in vitro studies model.Respectively, effects of E2or/and P4to this model, secretory activity of TNF-α, TGF-β andLIF were determined by ELISA, and expression level of progesterone receptor(PR) andestrogen receptor(ER) were determined by immunofluorescence technique.The resultsshowed as follows:1. Secretory activity of endometrial cells effected by sex hormones, result showed: whenEEC cultured alone, added E2and P4, alone or together, monolayer EEC were enhancedmarkedly, TGF-β secretory activity of both EEC apical and basolateral(P<0.05), andsecretion of TNF-α on EEC apical(P<0.05), but not significant on basolateral of TNF-α.When in coculture of ESC-EEC system, compared with EEC cultured alone, secretion ofTNF-α, TGF-β and LIF all are higher. On the other hand, compared with no hormone controlgroup, when added alone or together E2and P4, both apical and basolateral of TNF-αsecretion were repressed significantly, as well as TGF-β on EEC apical(P<0.05). While,when EEC cultured alone or together with ESC, added E2and P4, secretion of LIF notsignificant effect.2. Expression of hormone receptor on endometrial cells effected by sex hormones, result showed: sex hormones E2or/and P4added when EEC cultured alone, the expression of PRand ER were differently regulated. The effect was not significant when added E2alone, but P4alone or together with E2, were all repressed significantly about expression of PR and ER(P<0.05), especially P4alone. When the sex hormones in coculture model together with ESCculture medium, effects of PR and ER expression on EEC were similar to EEC culturedalone.
