| Objectives:To provide experimental evidence for clinical therapy of endocrin disorder after stress by investigating the influnce of non-trumatic stress to the hypothalamus-pitui-tary gland-adrenal gland axis.Methods:Selected health New Zealand big-eared white rabbit, all of the adult males, were divided into control group, bonded group, electroacupuncture-nonacupoint group, electroacupuncture-acupoint group randomly. Rabbits from each group were put to death after 12h,24h and 48h respectively. All rabbits were bonded supinely on self-made board except the rabbits from control group. Rabbits from electroacupunc-ture-acupoint group were punctuered by acupuncture with the depth of 0.3 to 0.5cm at 'Sanyinjiao' on one side, and those from electroacupuncture-nonacupoint group were punctuered by acupuncture as the same depth as the former group at the site 0.5cm apart from 'Sanyinjiao'. The frequency of stimulus of the two groups were 2 Hz, and stimulus intensity were no more than 0.6 mA, as strong as jitter's occurrence of the local skin and muscle. The stimulus of acupuncture lasted for 10 min each time and it was repeated every 20 min. The whole time of stimulus was 6 hours. Blood were collected from vein of all rabbits 3 hours,6 hours after the stimulus and before the rabbits were put to death.Serum were collected through centrifugate and conserved for test. ELISA technique were used to detect the concentration of CRH, ACTH, GHRH, GH, TRH and TSH in serum. After putting the rabbit to death, hypothalamus and pituitary gland were made into parafin section, Underwent HE dyeing, the latter were used for apoptosis and pathological detection.Results:the change of the hormone concentration in serum were monitored dynamically after stimulating the rabbits. The level of CRH, ACTH, GHRH, GH, TSH in serum were higher than that of control group significantly. After stress, besides the control group, the level of ACTH and GH were maintained at a higher and steady level, and the level of CRH, GHRH and TSH of experimental group fluctuated significantly at 6 to 12 hours after their increase which may correlated with multiple-path negtive feedback. After stress, compared with the control group, the level of TRH of experimental group increased slightly whereas that of the electroacupuncture-nonacupoint group and electroacupuncture-acupoint group fluctuated significantly 12 hours after stimulus.The level of all the 6 hormones of the electroacupuncture- nonacupoint group and electroacupuncture-acupoint group had certain difference. Brown apoptosis cells can be seen in hypothalamus and pituitary gland of experimental group in pathological detection, which suggested that stress stimulus could hurt hypothalamus and pituitary gland directly.Conclusions:1. The change of neuro-endocrine was significant in acute phase after stress stimulus. The level of CRH, ACTH, GHRH, GH and TSH increased immediately after stimulus except TRH.2. The level of CRH, GHRH, TRH and TSH fluctuated significantly whereas that of ACTH and GH maintained at a higher and steady level.3. The increased level of hormone in acute phase took long time to recover normal or a slight higher level.4. Apoptosis cell could be detected in hypothalamus and pituitary gland after stress. The number of apoptosis cell increased with the intensity of stress stimulus. Abvious stronger stress stimulus could hurt hypothalumus and pituitary gland directly which may became the pathological foundament of PTHP.5. PTHP influenced the prognosis of patients, and its incidence rate was higher one year after TBI. The diagnosis of PTHP and hormone replacement therepy need further reserch and guideline. |
