The marchantiaceae plants are well-known traditional Chinese medicinal herbs that are used extensively to treat skin burns, pneumonia and so on. There are many terpeniods and aromatic compounds in marchantia plants, some of which show good biological activities. The previous studies showed that the biological activities were related to flavonoids, triterpenoids and polysaccharide. In this paper, the extraction, determination, separation, purification of flavonoids and its antioxidant activities were systematicly studied, which will provide an experimental basis and theoretical basis for the exploitation of bioactive components of Marchantia paleacea. The main contents and conclusions are summarized as follows:1. Extraction of total flavones. Both ultrasonic-assisted extraction and microwave-assisted extraction were systematically performed to obtain a better extraction rate. Several factors influencing extraction rate of Marchantia paleacea were investigated, such as the concentration of ethanol, liquid feed ratio, temperature, time, the power of ultrasonic and microwave. Comparing extraction rate of flavonoids in Marchantia paleacea with different methods by single factor experiments, the better result was achieved by ultrasonic-assisted extraction and the rate of total flavonoids reached up to 1.35%.2. Determination of flavonoids in Marchantia paleacea by high performance liquid chromatography. A method was developed for simultaneous determination of four flavonoids in marchantia paleacea by SPE-HPLC. The sample was extracted by 70% ethanol and pretreatmented by the solid phase extraction with C18 as stationary phase, and separated and determined with methanol-acetonitrile-0.5%HAC solution (150:100:150, v/v) as mobile phase and a Kromasil C18 (150mm×4.6mm,5μm) as chromatographic column. All analytes showed good linearity in the test ranges and the recoveries were in the range of 89.70%-97.95%. Satisfactory precisions were obtained with relative standard deviations less than 4%. The method was proved to be simple, rapid, accurate, repeatable, and can be used for the quality control of flavonoids in Marchantia paleacea.3. Separation and identification of flavones monomer. The flavonoids compounds from the ethyl acetate extracts and n-butanol extracts of Marchantia paleacea were separated by HSCCC and their structures were characterized by means of IR, MS and HNMR. There monomers obtained were:quercetin, apigenin, apigenin-7-O-β-D-glucoside.4. The vitro activites of different extracts from Marchantia paleacea were evaluated. The results showed that there were different scavenging activities on superoxide anion radicals, DPPH radicals and hydroxyl radicals with different extracts, and their scavenging abilities enhanced with the increase of the concentration.
