Objective:To explore the relationship between the hypermethylation status of p16 and FHIT genes in Hazak and Han patients with esophageal carcinoma and the develop-ment of esophageal cancer, and analyse the prognosis.Methods:The hypermethylation of the p16 and FHIT genes were detected by methylation specific polymerase chain reaction (MSP) in 71 cases of Hazak and Han patients' esophageal cancer tissue. And then to analyse the relationship among the methylation status,age, gender,lymph nodemetastasis, Differentiation,clinical staging and the prognosis. Results:In 36 cases of Hazak patients with esophageal cancer,the p16 gene methylation in esophageal squamous cell carcinoma tissue samples and adjacent tissue samples were 44.4% and 13.9%; the FHIT gene methylation were 58.3% and 19.4%.In 35 cases of Han patients,the p16 gene methylation in esophageal squamous cell carcinoma tissue samples and adjacent tissue samples were 42.9% and 14.3%; the FHIT gene methylation were 51.4% and 14.3%. The difference of p16 gene methylation between kazak and Han patients was statistically significant (χ2=7.360,P=0.007;χ2=7.000,P=0.008),and the difference of FHIT gene methylation was statistically significant too (delete) (χ2=11.455,P=0.001).The difference between kazak and han nationalities in p16 gene methylation was not statistically significant (χ2=0.018,P=0.893;χ2=0.002,P=0.962),and the difference of FHIT gene methylation was not statistically significant too (delete)(χ2=0.342,P=0.559;χ2=0.336,P=0.562).Multivariate Prognostic analysis showed that the depth of tumor invasion,pathological stage, lymph node metastasis of the numbers of esophageal cancer area were independent prognostic impact of risk factors and gender was a protective factor for prognosis of esophageal cancer. Conclusions:The p16 and FHIT genes methylation were closely related to the occurrence of esophageal cancer in Kazak and Han patients in Xinjiang.
