Objective:This thesis was to discuss the effects of neuropeptide Y(NPY) and tyrosine hydroxylase(TH) in the occurrence and development of hypertension by observing the changes of neuropeptide Y(NPY), tyrosine hydroxylase(TH) and ultramicrostructure in superior cervical ganglion and adrenal medulla of spontaneous hypertensive rat.Method:We selected respectively 28 spontaneously hypertensive rats(SHR) and Wistar-Kyoto (WKY) rats in random.①The immunohistochemical PV-9000 method was used to examine the expression of NPY and TH in superior cervical ganglion and adrenal medulla of SHR and WKY rats. The positive signals were analysed by Image-Pro Plus6.0 analysis system.②Real-time PCR was used to examine the mRNA of NPY and TH in superior cervical ganglion and adrenal medulla of SHR and WKY rats.③The transmission electron microscopic was used to observe the changes of ultramicrostructure in superior cervical ganglion and adrenal medulla of each groups.Result:①Changes of the blood pressure:The blood pressure of SHR was obviously higher than this of the WKY (P<0.05).②The results of the immunohistochemistry:We can observe the NPY and TH immunopositive cells in superior cervical ganglion and adrenal medulla of each groups. The expression of NPY and TH protein in superior cervical ganglion of SHR was increased than those in WKY (P<0.05), the expression of NPY protein in adrenal medulla of SHR was much more than those in WKY (P<0.05), but the expression of TH protein in adrenal medulla of SHR has decreased (P<0.05).③The results of Real-time PCR:The result of Real-timePCR and that of immunohistochemistry corresponded with each other.④Changes of the ultrastructure:The number of rough endoplasmic reticulum, chondriosome, cytolysosome and secretory granules in superior cervical ganglion and adrenal medulla of SHR group was much more than WKY group. The structures of rough endoplasmic reticulum and chondriosome were destroyed.Conclusion:The expression of NPY, TH protein and mRNA were increased in superior cervical ganglion of adult SHR (hypertension formed). The expression of NPY protein and mRNA in adrenal medulla of SHR was increased, but the expression of TH protein and mRNA was decreased. There were some changes of ultramicrostructure in superior cervical ganglion and adrenal medulla of SHR. The upregulation of these factors might be closely related to the occurrence and development of hypertension. So we inferred that they may participate in the occurrence of hypertension by some complex mechanisms.
