ObjectiveTumor cell proliferation and angiogenesis growth play a key role in development of uterine leiomyoma.IGF-Ⅱcould activate RAS/RAF/MAPK and PI3K/AKT/PKB signal transduction pathway, and accelerate proliferation of cell.TSP-Ⅰis important inhibits angiogenesis factor.To study the expression of IGF-Ⅱ,TSP-Ⅰand contents of sex hormone in leiomyomata and adjacent normal myometrium, discuss their inter-correlations combining with occurrence and development of uterine leiomyoma. To demonstrate the pathogenesy of uterine leiomyoma and provide theoretical basis and experimental evidence for the non-surgical treatment of uterine leiomyomata.Materials and Methods1.30 patients with uterine leiomyoma from the third affiliated hospital of ZhengZhou University from December 2009 to April 2010 were enrolled.The average age is 42.58 (41.8±8.0).Obtain specimens via informed consent of patients and medical sector. In the following criteria to filter:(1) Operation time was the eighth~fourteenth day of menstrual cycle. (2) All specimen confirmed by post-surgical Pathological diagnosis. (3) All patients had regular menstruation Cycle, and had not used sex hormone drugs in the last three months.2. Thirty patients with uterine leiomyoma were enrolled in this study.Two pieces of uterine leiomyomata tissue and two pieces of djacent normal myometrium tissue which are over 2.5cm far away were cut as the study object in the mid of operation.A piece of tissue for determing E2 and P contents, another tissue for determing the expression TSP-Ⅰand IGF-Ⅱin uterine leiomyomata tissue and djacent normal myometrium tissue by immunohistochemistry.3. Statistical analysis was performed with SPSS 16.0 software. Paired t test was used to evaluate differences between two matched groups.correlation analysis using spearman rank correlation analysis. The level of significance was set at a=0.05.Results1. The TSP-Ⅰexpression is both in uterine leiomyoma tissue and adjacent normal myometrium tissue. The TSP-Ⅰexpression in uterine leiomyoma tissue (112.42±5.50) is lower than that in adjacent normal myometrium tissue (155.77±9.57). there was statistical significance (P<0.05)2. The IGF-Ⅱexpression is both in uterine leiomyoma tissue and adjacent normal myometrium tissue. The IGF-Ⅱexpression in uterine leiomyoma tissue (154.96±4.52) is higher than that in adjacent normal myometrium tissue (111.86±5.85). there was statistical significance (P<0.05)3. The contents of E2 in uterine leiomyoma tissue(150.43±53.96) is higher than that in adjacent normal myometrium tissue (80.75±20.29). there was statistical significance (P<0.05)4. The contents of P in uterine leiomyoma tissue (2.94±1.12) is higher than that in adjacent normal myometrium tissue (2.78±1.36). but did not reach statistical significance (P>0.05)5. The correlation is posive between the expression TSP-Ⅰand the contents of E2 in uterine leiomyoma tissue (r=0.668, P<0.05)6. The correlation is negative between the expression TSP-Ⅰand the contents of E2 in uterine leiomyoma tissue (r=-0.760, P<0.05)Conclusions1. Lower expression of TSP-Ⅰcan improve the development of uterine leiomyoma through increase vascular formation.2. Higher expression of IGF-Ⅱcan improve the development of uterine leiomyoma through promote the proliferation of leiomyoma cells.3. We speculate that TSP-Ⅰand IGF-Ⅱmight be important regulator of the sex hormone. When estrogen binds to estrogen receptor, it will promote the expression of IGF-Ⅱand inhibition the expression of TSP-Ⅰin uterine leiomyoma tissue.Higher expression of IGF-Ⅱand lower expression of TSP-Ⅰin uterine leiomyoma tissue can improve the development of uterine leiomyoma through promote the proliferation of leiomyoma cells and increase vascular formation.
