ObjectiveBy scanning electron microscopy and transmission electron microscopy, morphological structure of bone collagen of Osteogenesis Imperfecta patients and of normal persons were observed and analyzed respectively. And then find the difference between them, analyze the significance of the difference. Discuss the mechanisms of bone fragility with Osteogenesis Imperfecta.MethodsThe bone samples with osteogenesis imperfecta as the experimental group were selected from October 2008 to November 2010 in our hospital, the samples with dislocation of the hip bone which were examined and confirmed to be no other skeletal diseases were choosed to be control group.and it had the same standard with the experimental group. The number of samples of the two group is 10.Specimen collection and the use of it had received the informed consent of the patients or legal guardians, and approved to our hospital ethics committee. All the specimens were taken from the iliac crest bone. After specimen collection, fully demineralized, and then use 0.9% NaCl or 0.1 phosphate buffer solution to wash attachments of the specimen surface, immediately placed in 2.5% glutaraldehyde (4℃constant temperature, over 2 hours) to make electron microscope specimens. The sample preparation methods of the two groups are identical. By scanning electron microscopy and transmission electron microscopy, the experimental group and control group were observed, the morphological structure of bone collagen was compared, and then concluded differences between them.ResultsScanning electron microscopy observations:1) Osteogenesis imperfecta patients with cancellous bone trabecular structure of bone collagen disorder, discontinuity, poor integrity, loss of normal morphology.2) Patients with osteogenesis imperfecta,the diameter of collagen fibers reduced, arranged in disorder, to be inconsistent, messy cross-linked with each other, lose their law.3) Patients with osteogenesis imperfecta, collagen fibers can not be, gathered into orderly, regular shaped collagen fiber bundles.Transmission electron microscopy observations:1) Patients with osteogenesis imperfecta, collagen fibers in the cross section lost the normal circular contour,and the surrounding structure was fuzzy.2) Patients with osteogenesis imperfecta, collagen fibers in longitudinal section, lost the normal cord-like shape and clear contour, the structure cannot be recognized clearly.Conclusions1) Patients with osteogenesis imperfecta, collagen fibers and collagen fibers bundles dispaly abnormal morphological structures.2) The abnormal collagen fibers and collagen fibers bundles, resulted in that inorganic hydroxyapatite crystals in bone were not depositionde normally in the organic matter, which maked the occurrence of osteogenesis imperfecta bone mineralization or mineral deficiency. It may lead to decrease the strength and increase the fragility of bones of osteogenesis imperfecta.
