Investigation Of Annonaceous Acetogenins On The Inhibiting Effect Of Lung Cancer And Its Mechanisms

Composed of two parts which include theoretical and experimental research,it discus about the Effect and its Mechanisms of annonaceous acetogenins on inhibiting Proliferation of the hu-man lung cancer cell.Theoretical study involves the clinical research of Western medicine and Chinese medicine on the lung cancer in recent years.Then,from the theory of TCM, on the basis of investigation and a large number of studys,We synthesized the clinical experience of instructor and academic think-ing and discussed the the evidence of "Jiedu"(detoxification and dispersing accumulation). Ex-pounded the theory that"Aidu"(poisonous pathogenic factor) is the etiological factor and pathol-ogic product through the whole course of lung cancer. Ancient literature recorded that the annon-naceous acetogenins can be insecticide^ poison for fish and abortifacient,also can treat rheumatis-m,malaria,measles and acariasis.Modern medicine confirms that annonaceous acetogenins can inhibit human cancer cells,parasite,malaria and T cell.To study the condition and mechanism of action of treatment of lung cancer induced by an-nonaceous acetogenins, we conducted a number of experiments.The first investigation of annona-ceous acetogenins on the anti-proliferation of lung cancer and its mechanisms.Methods:build a model of the solid tumor of transplanted cancer on mouse,observe the rate of the inhibition of tumor growth.The result of this experiment shows that the rates of the inhibition of tumor were 5.85%,-2.79%,5.07%respectively in the cases of 6.25mg,12.5mg/kg,25mg/kg. The result shows that annonaceous acetogenins has no inhibition on the growth of mouse transplanted cancer. The seond:Study on the effects of inhibiting of pomegranate annonaceous acetogenins on human lung cancer cell.Methods:(1) The MTT assay was used to detect the growth inhibition rates of A549cells and NCI-H460 cells at 72h which cultured with annonaceous acetogenins in different concentrations.(2) Flow cytometry was used to observe the apoptosis induced by annonaceous acetogenins in human lung cancer.Results:(1) Annonaceous acetogenins inhibited the proliferation of human lung cancer cell line A549 and NCI-H460.The different concentrat-ions of annonaceous acetogenins had cytotoxicity on lung cancer cells, which were dose-depen dent. (2)Cells were treated with annonaceous acetogenins induced apoptosis.Conclusions: Annonaceous acetogenins inhibit human lung cancer cell line A549 and NCI-H460 cell proli-feration, and can induce lung cancer cell lineA549 and NCI-H460 apoptosis.
