Objective:Colorectal carcinoma is one of the most common malignant tumor in our country, the morbidity rate and mortality rate are increasing year after year. The development of colorectal carcinoma not only refers to the activation of proto-oncogene and the in activation of anti-oncogene, but also correlates with other genes in recent years' studies. Mcm7 is a good indicator for reflecting the degree of cell proliferation sensitively; its expression is increasing gradually from premalignant lesion to canceration. hRad17 is an early replication checkpoint protein which sensing DNA lesions and halting progression of replication fork in cell cycle.It plays an important role in insuring the integrality and stability of DNA. Its functional defect and high expression may lead to a predisposition to cancer and may have a relationship with carcinogenesis and progression of carcinoma. To investigate the expression of Mcm7 and hRad17 in colorectal carcinoma and study actions in carcinogenesis and progression of carcinoma and evaluate their relationship.Methods:Immunohistochemical technique (s-p method) was used to examine the expression of Mcm7 and hRad17 in 60 cases colorectal carcinoma and 28 cases adjacent normal tissues. The concentration of both Mcm7 and Rad17 were 1:600. We carry out experiments according to the specification. The negative control group was added PBS buffer instead of antibody.Results:The expressions of Mcm7 in colorectal carcinoma:The positive expression of Mcm7 was detected in cell nucleus of cancer tissue. The positive rate of Mcm7 in colorectal carcinoma tissues was 76.67%,but in adjacent normal tissues was 0(0/28) (χ2= 44.98, P<0.05).The positive labeling indexes of hRad17 (76.67%),was notably higher than that in adjacent normal tissues 17.86%, (χ2=22.295) P<0.05) Expression differences of Mcm7 in cancer tissues and adjacent normal tissues have remarkable statistics significance.(2) The expressions of hRad17 in colorectal carcinoma:The positive expression of hRad17 was detected in cell nucleus of cancer tissue. The positive labeling indexes of hRad17 (76.67%)was notably higher than that in adjacent normal tissues 17.86%, (χ2 =22.295) P<0.05). Expression differences of hRad17 in cancer tissues and adjacent normal tissues have remarkable statistics significance.(3)Expression of Mcm7 and clinical pathologic parameters in colorectal carcinoma: The expression of Mcm7 has a relationship with differentiation grade of cancerous cell. Positive incidence of highly differentiated group was 16.67%, positive incidence of moderately differentiated group was 76.00%. Positive incidence of poorly differentiated group was 89.66%. The difference among three groups have statistics significance. (χ2=14.816 P<0.05).At the same times, the positive correlation was existed between Mcm7 and hRad17's expression in colorectal carcinoma. Expression of Mcm7 was not related to age,sex,tumor size and histological type of tumor (P>0.05)(4) Expression of hRadl7 and clinical pathologic parameters in colorectal carcinoma:It has a relationship between hRad17 and lymph node metastasis. Expression rate of hRad17 was 87.18% in 39 cases of lymph node metastasis group,(34/39).Expression rate of rad17was 52.38% in 21 cases without lymph node metastasis group(11\21), (χ2=8.82. P<0.05). Expression of hRad17 was not related to age,sex,tumor size and histological type of tumor (P>0.05)The Spearman's rank correlation analysis indicated that Mcm7 has a positive correlation with hRad17 (p<0.05)Conclusion:(1) Mcm7 was a good marker for cell proliferation. It may be an early factor for the progression of tumor. Determination of Mcm7 genes expression in colorectal carcinoma tissue can be of use in deciding the malignant degree of the tumor, the therapeutic effect and the recurrence after curative surgery.(2) hRadl7 is a new drug-resistant and metastasis gene. Genetic checking of hRad17 way be able to provide colorectal carcinoma patients rational treatment programs.(3) Mcm7combine with hRad17 functionally. The interaction between Mcm7 and hRad17 may play a role in carcinogenesis and progression of carcinoma. Immunohistochemical technique was used to prove that correlation exist between the two genes in this article.
