Objective:Transfect the siRNAwhat could inhibit the expression of ubiquitin-related protein FAT 10 into Hep3B and MHCC-97h cells by LipofectamineTM 2000.Explore the influence of Chemotherapy sensitivity for the hepatoma carcinoma cells MHCC-97h and Hep3B after FAT10 interfered. And provide theoretical basis for the further research of the multimodality therapy of HCC.Methods:1.The bunches of cells divided into six groups:Norma,FAT10 interfered,cis-platinum,FAT10 interfered and cis-platinum,transfect the siRNA into Hep3B and MHCC-97h cells by LipofectamineTM 2000.Measuring the growth curve of each group.2. The Expression changes of mRNA of bcl-2 and bax were estimated by RT-PCR, western-blot was used to detect the protein of bcl-2 and bax in every group.3. the cell growth cycles and apoptosis were analyzed by the folwcytometry.Results:The FAT 10 interference demonstrated by the result of RT-PCR and Western blot. The results of RT-PCR and Western-Blot shows that the expression of protein of Bcl-2 reduced while Bax increased. The growth cycle of group that FAT10 interfered has been suppressed,and the group that FAT 10 interfered and with cis-platinum is more obvious,and mainly in G0/G1 phase,and the apoptosis index increased.Conclusion:The chemotherapy sensitivity of HCC has been enhanced after FAT10 interfered.May provide theoretical basis for the further research of the multimodality therapy of HCC.
