Experimental Study Of Antivirus Effect Of Gentiana Veitchiorum,alstonia Scholaris, Againest Rsv In Hep-2 Cells

Objectives:To study the antiviral effects and mechanisms of Gentiana veitchiorum,Alstonia Scholaris,Cytisine in vitro and in mice infected with RSV.Methods:1. The effect of Gentiana veitchiorum,Alstonia Scholaris,Cytisine on hep-2 cells infected with Respiratory Syncytial Virus ,including the effect of inhibiting virus replicatin,direct killing and Lesions blocking.were observed by the cytopathic effect of (CPE) and MTT colorimetry methods ,the toxic effect of all of Chinese drugs is also observed.2. After establishing the mice models infected with RSV,the effect of Gentiana veitchiorum,Alstonia Scholaris on anti-lung infectious were observed by HE staining method and immunofluorescence method. Results:1. The result of complement dependent cytotoxicity is Gentiana veitchiorum 6.25mg/ml,Gentiana veitchiorum decoction, n-butyl extract 1700mg/ml,ethyl acetate extract 19.1mg/ml,chloroform extract 7.5mg/ml,Alstonia Scholaris 12.5mg/ml. In vitro test.Compared with the virus control group, all of the Gentiana veitchiorum extract can inhibit the rsv replication in Dilution 32 times.All of the Chinese medicine extracts have no effect in direct killing and blocking virus attack on anti-RSV. all of the Chinese medicine extracts maybe have a littie effect on blocking virus attack..2. The pathological changes of the lung dyed with HE stain were observed through light microscope in vivo experimental animal model,The results under: virus control group alveolar space and a large number of small airway inflammatory cell infiltration, edema and bleeding obvious,the alveolar septa was widened. Compared with virus control group, the infiltration of inflammatory cells were decreased under the electro microscope in Gentiana veitchiorum group and Alstonia Scholaris group.Conclusion:1. All of the Gentiana veitchiorum extracts have the function of inhibiting Hep-2 cell lesion after RSV virus infection.2. Gentiana veitchiorum decoction and Alstonia scholaris decoction be able to decrease the lung inflammation of RSV infection.
