ObjectivesOral and maxillofacial malignant tumor accounted for about 3% of the body, and approximately 32.08% in the oral and maxillofacial tumors, cysts and tumor-like lesions. Although the percentage is not high, because of specific anatomic site, involving beautiful and important physiological functions, and therefore the influence of patient's body and mental can not be ignored. That disorder and endless growth, invasion and metastasis are three main biological characteristics of the malignant tumor. Local invase is the fundamental difference between carcinoma and non carcinoma. According to these characteristics, the present main treatment includ surgery, chemotherapy and radiotherapy, and the new biological therapy and rehabilitation therapy, and so on. Chemical medication, as important means of clinical tumor therapy, has been recognized by basic researchers and clinical doctors, but its poison effect can't be ignored, like myelosuppression and gastrointestinal reaction which make patients physical and mental bear great pains. Therefore, it appears especially important to find new low-toxic and effective antitumor drugs in the treatment of tumor.With the development of the genetics, immunology and molecular biology, the research in cancer drugs has already developed from the cytotoxic to new antitumor drugs, such as growth factor inhibitors, angiogenesis inhibitors, biological response modifier, tumor drug-resistant reversal agents, genetic engineering drugs and so on. Indoles acetic acid (IAA) is a kind of auxin which plays a key role in plant cell division, growth and differentiation. Horseradish peroxidase (HRP) is a plant hormone containing peroxidase, which can oxidize a series of substrates including indole acetic acid. HRP was used to transform the prodrug IAA and other indole analogues to toxic drugs to kill cells in vivo. IAA has a high tolerance in the human body and can be monoamine oxidase in mammalian metabolism, without being oxidized by peroxidase. Only in the presence of HRP, will IAA be oxidized into toxic substances, and thus produce toxic effects on the surrounding tissue cells.Oral and maxillofacial squamous cell carcinoma(OSCC) which is one of the highest morbidity and mortality of tumor comes from epithelial malignancies and accounts for more than 90% of them. Tca8113 tongue squamous cell carcinoma has already maturely plant. The experiment study how HRP/IAA therapy the tumor, and then explore the therapeutic mechanism, and provides the experiment basis for clinical application.MethodsEstablish the nude mice model, and divided into three groups with eight mice in each group, such as control group, positive control group and experimental group. The experiment time is total twenty-one days. What should be observed from the third day is the tumor volume, weight and vital sighs of the nude mice. Killing the mice at the twenty-three day, and taking out the tumor, weighting and calculating inhibition rate. After all, detecting the expression of PCNA and Bcl-2 by immunohistochemistry. Dissecting out the heart, liver, lung, spleen, kidney and testis from the nude mice, and observing the organ morphology and understanding the toxicity of drugs by using HE staining.ResultsThe inhibition rate of positive control and experimental was 56.02%,49.21%. They are statistically significant comparing with the control group(P<0.05); There is no significant difference between the control group and experimental group in weight of the mice(P>0.05). There is no obvious poison effect on the visceras through HE staining; There is significant difference comparing the positive control and experimental with control group by observe the expression of PCNA and Bcl-2 (P<0.05), but no difference between the positive control and experimental(P>0.05). Conclusions1. The mechanism of HRP/IAA antitumor may be promote tumor cell apoptosis, so as to achieve antitumor effect and inhibit tumor growth.2. HRP/IAA has antitumor effect on the Tca-8113 tumor-burdened nude mice,and can inhibit tumor growth.3.
