Backgroud:Non-alcoholic fatty liver (NAFLD) is one of common chronic diseases, existing in many countries. The overall incidence of NAFLD growed rapidly in the past fifteen years. The disease eventually develop cirrhosis or liver cancer, which makes the prognosis more serious. As the pathogenesis of NAFLD gradually study, the deposition of fat in the development and progression of disease played a key role. And adiponectin (adiponectin) and liver specific receptor (adipoR2) plays a key role in the regulation of lipid metabolism and deposition in the liver. For this situation, we study the effective components of Chinese medicine and found that emodin (emodin), mainly in the rhubarb, fleece-flower root, Polygonum and other Chinese medicinal herbs, has some effects on fat liver disease. Mechanism of Emodin on NAFLD should be investigated.Objective:To observe the effects and mechanism of emodin treating hepatic lipid deposition in mice.Methods:C57BL6 black mice feeding high fat diet for 12 weeks. The establishment of NAFLD model were randomly divided into model group, emodin group and the rosiglitazone group. drug intervention, once a day respectively. the model group (5% DMSO20mg/kg), emodin (20mg/kg), rosiglitazone (10mg/kg). After 4 weeks of drug intervention, the body weight, liver weight, liver index, liver triglyceride, liver pathology and liver cells adiponectin receptor R2 expression were observed.Results:C57BL6 mice received high fat diet after 12 weeks, NAFLDA model were successfully established. After drug intervention, body weight and liver weight, decreased liver TG levels were more reduced mice in the emodin group, compared with those in model group and the rosiglitazone group. Liver pathology in mice suggest that the liver cell structural integrity of liver cell is improved, lipid droplets in the cytoplasm is essentially deceased. Liver adiponectin receptor expression R2 in emodin group increased slightly compared with model group,while the rosiglitazone group were not significantly different.Conclusion:emodin can reduce body weight, lower levels of TG within the liver cells and reduce fat deposition within the liver, significantly improve the organizational structure of mouse liver. The mechanism of emodin treatment of NAFLD may lie in the increasement of adiponectin receptor R2 expression.
