| [background] Peritoneal dialysis is one of the renal replacement therapies for end-stage kidney patient. Peritonitis remains a leading complication of the peritoneal dialysis.Previous studies showed that conventional high glucose dialysate and peritonitis can induce expression of inflammation factors such as IL-6 and IL-8 which are involved in peritoneal fibrosis.[objective] to investigate the effect of ligustrazine on IL-6 and IL-8 expression induced by high glucose or TNF-a(tumor necrosis factor-a)in cultured human peritoneal mesothelial cells (HPMCs).[methods] HPMCs were isolated from human omenta by trypsin digestion method and subcultured. After synchronization of cells growth, HPMCs were exposed to normal control group, high glucose group and high glucose plus low-, medium-and high-dose ligustrazine (10, 20 and 40 mg/L ligustrazine respectively) groups. HPMCs were also exposed to normal control group, TNF-a group and TNF-a plus low-, medium-and high-dose ligustrazine (10,20 and 40 mg/L ligustrazine respectively) groups. Semi-quantitative reverse transcriptive-polymerase chain reaction (RT-PCR) was used to detect the expressions of IL-6 and IL-8 mRNAs in HPMCs. Protein levels of IL-6 and IL-8 in the culture medium supernatants were determined by enzyme linked immunosorbent assay (ELISA). Cells protein concentration measured by trace BCA method was to correct the ELISA assay results.[results] Compared with the control group, high glucose can significantly reduce HPMCs activity (P<0.01), and significantly increase the expression of IL-6 and IL-8 (P<0.01); after ligustrazine intervention protein levels of IL-6 and IL-8 were significantly decreased (P<0.05), compared with high glucose group. IL-6 mRNA and IL-8 mRNA expression were significantly reduced (P<0.05). Compared with control group, TNF-a can significantly reduce HPMCs activity (P<0.01), and highly increase the expression of IL-6 and IL-8(P<0.01); After ligustrazine intervention, protein levels of IL-6 and IL-8 were significantly decreased (P<0.05, P<0.01), and IL-6 mRNA and IL-8 mRNA expression were significantly reduced (P<0.05), compared with TNF-a group.[conclusion] The present data demonstrated that ligustrazine does not only decrease high glucose-induced IL-6 and IL-8 synthesis, but also decrease TNF-a-induced IL-6 and IL-8 synthesis.
