Object:Observe the effects of suppressing the multiplication of person ovarian cancer cell line SKOV3 under different density of Brucea Javanica Oil Emulsion Study the possible mechanism and action principle.Methods:Use MTT to test after deal with the person ovarian cancer cell SKOV3 under different density(1.25ml/L,2.5ml/L,5.0ml/L,10ml/L,20ml/L) of Brucea Javanica Oil Emulsion, the multiplication situation of the cells. Calculate the suppress rate (%)= (1-experimental group extinction value/ control group extinction value) x100%. Repeat the experiment steps three times. Take the average value of the three experiments. Take the medicine density as the abscissa and take the cell suppress rate as Y-coordinate. Use Electron microscope to observe the Morphological change of cells before and after medicine. The experiment divides into the blank control group and the experimental group. The experimental group establishes a branch four densities:5.0ml/L,10ml/L,20ml/L Brucea Javanica Oil Emulsion. Cultivate them 48 hours after medicine and collect cells, prepare to observe with Electron microscope.Results:1 Brucea Javanica Oil Emulsion in the range of 1.25-20ml/L has inhibitory action to the multiplication of person ovarian cancer cell SKOV3. Its intensity will increase with the time and concentration of drug increased and become time-dose dependent.Compared with the control group.the difference was significantly significant(P<0.05)2 The result of the morphological examination of ovarian cancer cells skov3 after treatment 48 hours by Brucea Javanica Oil Emulsion shows following with the density of the drug increased, the cell diameter was decreased more obviously. Apprearing necrotic cellular debris, Microvillus and false feet attenuated.Nuclear membrane structure uncleared.It can observe Choromatin condensation or margination. Also the autophagy was more obvious when the concentration of the drug increased.Conclusion:1 Brucea Javanica Oil Emulsion has the function which to person ovary cancer cell SKOV3 the obvious multiplication suppresses and induces apoptosis. The apoptotic rate of cell was of time and dose dependent. It is difference between every time group (P<0.05).It is difference between every density group (P<0.05)The effect of Brucea Javanica Oil Emulsion to ovarian cancer cells skov3 after 48h, By Electron microscope showed cells in every treatment groups appeared morphological change at different levels in different density.
