Objective:To evaluate the effect of insulin and trimetazidine on myocardial function and perfusion of diabetic cardiomyopathy in rats with velocity vector imaging and myocardial contrast echocardiography.Methods:Eighty male SD rats were divided into control group and model group randomly. The models were established by intraperitoneal injection of streptozotocin, and then divided into three groups: diabetic mellitus group (DM, n=20), insulin treated group(INS, n=20) and trimetazidine treated group(TMZ, n=20). INS group were given insulin hypodermic injection at dose of 6 U/kg and TMZ group were given trimetazidine 10mg/kg by gastric lavage daily ever since DM models were established. After 8 and 12 weeks, each rat was performed with echocardiography to evaluate the VVI and MCE parameters, after that, myocardium was collected for pathology and transmission electron microscope examination.Results:1.VVI parameters :After 8 weeks : Compared with control group, Vs,Vd,Sr,systolic and diastolic SRr,Sc,systolic and diastolic SRc were significantly lower in DM group(P?0.05), Vs,Vd,systolic and diastolic SRr,systolic and diastolic SRc were also lower in INS group and TMZ group(P?0.05; In contrast with DM group, all parameters in INS group and TMZ group were increased apart from Vs indicator ; There had no statistical difference between TMZ group and INS group(P?0.05).After 12 weeks: Compared with control group, Vs,Vd,Sr,systolic and diastolic SRr,Sc,systolic and diastolic SRc were significantly lower in DM group(P?0.05), systolic and diastolic SRr,systolic and diastolic SRc were also lower in INS group and TMZ group(P?0.05); Compared with DM group, all parameters in INS group and TMZ group were increased apart from diastolic SRr indicator; There had no statistical difference between TMZ group and INS group(P?0.05).2.MCE parameters:After 8 weeks: Compared to control group, A,β,A×βwere significantly lower in DM group, A×βwere decreased in INS group and TMZ group(P<0.05);Compared with DM group , A×βincreased in TMZ group(P<0.05); There had no statistical difference between TMZ group and INS group(P?0.05). After 12 weeks:Compared to control group, A,β,A×βwere significantly lower in DM group; parameters in INS group and TMZ group were increased when compared with DM group (P<0.05);There had no statistical difference between TMZ group and INS group(P?0.05).Conclusion:The myocardial function and perfusion in diabetic rats have significantly improved after treated by insulin and trimetazidine; SR and MBF parameter plays an important role in the assessment of curative effect and diagonosis about diabetic cardiomyopathy.
