Objective Observed proliferation and differentiation of dentate gyrus neural stem cells after intracerebral hemorrhage(ICH)in adult and aged rats. Discussion the variation of neural stem cells after ICH.Method 192 healthy Sprague-Dawley rats, 96 in adult rats and aged rats , either male or female. Each adult and aged rats were randomly divided into normal group(18 rats), sham operation group(12 rats)and ICH model group(3 d, 7 d, 14 d, 21 d, 28 d, 66rats). ICH model was established by injecting collagenaseⅦinto the brain of rats. The normal group and sham operation group were killed in 7d, ICH model groups were killed in 3 d, 7 d, 14 d, 21 d and 28 d. All rats were sacrificed before 24h intraperitoneal injection with Brdu to mark proliferating cells. Behavior was scored by Bederson score Standard. Detected number of proliferating cells labeled by immunohistochemistry. Observed differentiation of proliferating cells marked by double target Brdu/NeuN and Brdu/GFAP.Results Score of neurological behavior in rats of sham operation group and the normal group had no difference in postoperative, hemorrhage group were symptom hemorrhage lesions contralateral hemiplegia, hemorrhage group were symptom hemorrhage lesions contralateral hemiplegia over time, Neurological function deficit of hemorrhage group gradually recovered. Water content of cerebral hemorrhage reached a peak at 72h and then gradually decreased, edema significantly reduce after 7d, resultd has Significant difference (P<0.05), ICH group has a fast recovery at each time point. Immunoassay results suggesting, Brdu positive cells has a small amoun in normal group and sham operation group, cerebral hemorrhage Brdu positive cells in each group significantly increased than in normal group and sham operation group (P<0.01), 3d group increased significantly, 7d group after reaching its peak has decreased gradually ,28d group still higher than the normal group and sham operation group. On the normal group and sham operation group a little of BrdU positive cells were found on the hippocampus dentate gyrus of aged rats, there were no significant difference between the two groups(P>0.05). On the hemorrhage group the quantity of BrdU positive cells increased obviously, and there were significant difference on the aged rats compared with the normal group and sham operation group(P<0.01). Compared with the adult rats group, the quantity of BrdU positive cells decreased obviously on the aged rats(P<0.01). A little of BrdU/NeuN and BrdU/GFAP double-marked positive cells were found on the hippocampus dentate gyrus of normal adult and aged rats, and the number of positive cells on the adult group was more than the senile group. After ICH the number of double-marked positive cells increased obviously compared with the normal group(P<0.01), furthermore the positive cells of BrdU/NeuN on the adult rats was more than on the aged rats, and the positive cells of BrdU/ GFAP on the aged rats were more than on the adult rats.Conclusion 1.A small amounts of nerve stem cell proliferation on the hippocampus dentate gyrus of normal rats, and the reproductive activity of adult rats was stronger than aged rats. 2.The reproductive activity of nerve stem cell proliferation on the hippocampus dentate gyrus of rats was reinforced after ICH which reached the peak value on the 7d after ICH, and the reproductive activity of aged rats was lower than adult rats. 3.The neuron differentiation of nerve stem cells on the hippocampus dentate gyrus of rats after cerebral hemorrhage. Between them the positive cells of BrdU/NeuN on the adult rats were more than aged rats, and the positive cells of BrdU/GFAP on the adult rats were lower than aged rats. 4.It was an important factor for the age to influence the nerve stem cell proliferation and differentiation after ICH. With the increase of age, the activity of the nerve stem cell proliferation and differentiation after ICH.
