To investigate the genetic association between single nucleotide polymorphism of matrix metalloproteinase 3 gene and esophageal cancer susceptibility, three SNPs within upstream transcriptional regulatory regions were selected to be analyzed:—774rs522616(A/G),—1380rs632478(A/C),—1552rs494963(T/G).Extracted genome DNA from peripheral blood with proteinase K method; then all three selected SNP sites of genome DNA samples, which were from sporadic cases of esophageal cancer in Henan, sporadic cases of esophageal cancer in Hubei, migrants of the high incidence area in Henan and healthy controls in both Henan and Hubei, were genotyped by PCR-RFLP and analyzed comparatively. Finally, the results of genotyping were analyzed by SPSS.(1)—774rs522616(A/G)allele has three gene types: GG, GA and AA; there were significant differences between healthy controls in Hubei and sporadic cases of esophageal cancer in Henan in genotype GG and AA (p<0.01), sporadic cases of esophageal cancer in Hubei and in Henan in genotype GG(0.01<p<0.05), AA (p<0.01), migrants of the high incidence area in Henan and sporadic cases of esophageal cancer in Henan in genotype AG(0.01<p<0.05), AA (p<0.01), sporadic cases of esophageal cancer in Henan and healthy controls in Henan in genotype AG(0.01<p<0.05), AA (p<0.01). Moreover, there were only differences between healthy controls in Hubei and migrants of the high incidence area in Henan in genotype GG (0.01<p<0.05).(2)—1380rs632478(T/G)allele has three types: TT, TG and GG; there was only difference between healthy controls in Henan and sporadic cases of esophageal cancer in Henan in genotype CC (0.01<p<0.05). (3)—1552rs494963(T/G)allele has three types: TT, TG and GG; there was only difference between healthy controls in Henan and sporadic cases of esophageal cancer in Henan in genotype TG and GG (0.01<p<0.05).Single nucleotide polymorphisms at rs522616,rs632478 and rs494963 of transcriptional regulatory regions in human matrix metalloproteinase 3 gene apparently associated with esophageal cancer susceptibility.
