Objective To investigate the protective effects of different extract fractions from Panax japonicus C. A. Mey preconditioning on acute myocardial ischemia injury in rats induced by ligating of the left anterior descending branch (LAD).. To investigate the protective effects and the mechanism of total saponin from Panax japonicus C. A. Mey pretreating on acute myocardial ischemia injury induced by LAD in rats.Methods Experimental one: All rats were divided into sham group, model group and treatment group. The pretreatment group given the fractions of Panax japonicus C. A. Mey including ethyl acetate, n-butyl alcohol and water fractions respectively. After pretreating for 7 days, the rats'acute myocardial ischemia model was induced by ligating of the left anterior descending branch (LAD). The infarct size were assessed at 12h after LAD, the serum levels of superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), malondialdehyde (MDA), cardiac troponin I (cTnI)were also measure. Experimental two: All rats were divided into sham group, model group and high-dose,middle-dose and low-dose group of total saponin of Panax japonicus C. A. Mey. The pretreatment group was given total saponin of Panax japonicus C. A. Mey. After pretreating for 7 days, the rats'acute myocardial ischemia model was induced by LAD in rats. The infarct size and cardiac function was assessed at 12h after LAD, the serum levels of LDH, SOD, CAT and GSH-PX were also measured. The expression of bcl-2,bax,Mn-SOD and Cu/Zn-SOD was detected by Real-Time quantitative PCR.Results Experimental one: n-butyl alcohol fraction from Panax japonicus C. A. Mey obviously reduced infarct size in acute myocardial ischemia injury rats; improved the activities of SOD and GSH-PX, and MDA, cTnI content were decreased respectively, ethyl acetate and water fractions have no significant effect. Experimental two: In comparison with the model group, high-dose of total saponin from Panax japonicus C. A. Mey significantly improved cardiac function in acute myocardial ischemia injury rats. high-dose of total saponin from Panax japonicus C. A. Mey significantly released CK, middle-dose increased SOD and CAT. All total saponin improved GSH-PX. high-dose and middle-dose groups obviously improved heart Mitochondria, reduced infarct size. Also, all totlal groups obviously increased the rate of bcl-2/bax, middle-dose and high-dose group decreased the expression of caspase-3. middle-dose group increased the expression of Mn-SOD, these were no significant exchange of Cu/Zn-SOD in every group. Conclusion The results suggest that n-butyl alcohol fractions from Panax japonicus C. A. Mey pretreating has significantly protective effects on acute myocardial ischemia injury in rats, ethyl acetate and water fractions have no significant effect. Total saponin from Panax japonicus C. A. Mey pretreating provides a protective effect on acute myocardial ischemic injury, one of the mechanisms is that it inhibit apoptosis via antioxidation.
