Objective to investigate the effects of Fasudil, a Rho kinase inhibitor, on the expression of RhoA,Rock- and CD8 cells of brain,spinal cords;to explore the mechanism of Fasudil in the experimental autoimmune encephalomyelitis(EAE).Methods 90 Lewis rats were divided into three groups: Lewis rats control, EAE animal models and Fasudil-treated groups. The EAE model was established in Lewis rats by immunization with complete Freundps adjuvant (CFA) and Guinea pig spinal cord homogenate (GPSCH).The clinical scores and expression and cell count of RhoA,Rock-Ⅱand CD8 cells of brain,spinal cords.Results to establish the EAE animal models successfully. The results show that Fasudil decreased the development of EAE in Lewis. Compared with control group, the expression of RhoA﹑Rock-Ⅱand CD8 cells of brain, spinal cords were increased in the EAE models and Fasudil-treated. The expression of RhoA,Rock-Ⅱand CD8 cells were significant difference in the Fasudil-treated compared with the other groups(P<0.05). However, there was no significant difference between day 13 post-immunization (p.i) with day 35 postimmunization(P>0.05). There is not significant difference between Fasudil-treated 13 day and 35 day. Conclusions1. RhoA, Rock-Ⅱ, CD8~+ T cells in the pathogenesis and progression of EAE play an important role.2. RhoA, Rock-II and the expression of CD8~+ T cells in EAE model changes and development trend of single-phase line course.3. Fasudil by inhibiting the inflammatory response, the dual role of promoting nerve regeneration, reduce EAE symptoms and improve prognosis.
