Study A:Objective: Through eight weeks diet adjustment ,observed after short-term no weight-bearing exercise and diet ,the adaptation change expression of AMPKbeta2 protein in rat sketal muscle cells,further more from adaptability perspective,infer sports and diet to the changes of gene experession of AMPKbeta2.Methods: Adult male wistar rats 35 were randomly divided into groups, only 10 (K group), short-term the exercise group 10 only (E0 group), sprint group of 10 only (E group) and high sugar group low-glycemic load transfer ELHE group only (5); One K group, the E0 group and group E divided into high sugar group (KH group, EH0group, EHE group) and low-carbohydrate group (KL group, EL0 group, ELE group) 6 teams of 5 only. In addition, all high sugar ELHE group group were given high sugar food and drink, low-sugar 5% sugar cane sugar group were given food and distilled water drinkable; E0 group and group E all rats 30min to meet every adaptability swimming exciting and swimming, water for eight weeks, six days a week, every day,six hours of rest 45 minutes after swimming training, rest after 72 hours of E0 slaughtered; Group E begin loading in 72 childhood continue to 12% of swimming, terminate the sports standard: rats not being returned to the water sank to underwater 10s, remove and more than 2 minutes continue to rest, so repeatedly for three times sport immediately after exercise terminated after killing. After training gastrocnemius laterally take rats, usewestern-style imprinting white muscle of total protein and analyzed skeletal muscles in the beta 2 AMPK nucleoprotein expression level; And the determination of copper with sulfuric acid in oxygenated muscle glycogen storage changes.Results: In 8 weeks of aerobic workout, observed that speed-training compared with not speed-training,low-sugar training group sketal muscle cells AMPK beta 2 total protein expression in increased, whereas high sugar training group did not change, at the same time the concentration of glycogen in high sugar group is obviously higher than overall low-carbohydrate group; EL0 group than EH0group, ELE group than ELHE group, the concentration of glycogen is lower, skeletal muscle of total protein expression in beta 2 AMPK haven't changed, nucleoprotein is higer; ELE group than EL0group,EHE group than EH0 group, in skeletal muscle cell,the expression of beta2AMPK nucleoprotein significantly higher,the expression of total protein present decreasing trend,and the concentrationmuscle of glycogendecreased.Conclusions: (1) In skeletal muscle cells ,the concentration of muscle glycogen and the expression of AMPKbeta2 hanve Close relationship, low Concentrationof muscle glycogen promote AMPKbeta2 protein expression, high concentrations of muscle glycogen inhibition the expression of AMPK beta2; (2)no matter in high muscle glycogen or in low muscle glycogen and low muscle glycogen turn high muscle glycogen model, all find skeletal muscle cells AMPKbeta2 positioning change are corresponding to muscle glycogen concentration change.Study B:Objective: Through diet intervention, observed the relationship between the positioning and the expression of beta2AMPK protein and the concentrationof muscle glycogen before and after three days load swimming in rats muscle gastrocnemius.Form the signal from AMPK based energy metabolism, explore the ways of key enzymes AMPK speed movement and muscle,and the mechanism of action of glycogen for improving sports abilities and diabetes prevention on the theoretical basis of providing further developed new drugs for diabetes and provide new molecular targets, provides the basis.Methods: 27 adult male wistar rats,were randomly divided into quiet group only (group S 5 only), sports made module (E0 group of 10 only) and sprint group (group E,10 only) by divided into two groups, high sugar group(HE0, HE) and low-carbohydrate group (LE,LE0), four teams of five; and AICAR group (high/low carbohydrate, HEA, LEA one per group) of With high ratio of low-glycemic index foods that are high in sugar diet groups, have given high sugar food and 5% sugar water drinkable, a low-glycemic load diet group is assigned a low-glycemic index foods and distilled water drinkable; Group S give normal rat food and distilled water. Then a three-day drive method by every six hours and 45 minutes rest between the weight loading swimming movement, negative weight for rats 5% of their body weight, three days after training, E0 72 hours of rest; slaughter Group E begin loading in 72 childhood continue to 12% of swimming, terminate the sports standard: rats not being returned to the water sank to underwater 10s,remove and more than 2 minutes continue to rest, so repeatedly for three times sports after termination of sports training after take rats AICAR gastrocnemius laterally white muscle, 72 hours after the rest of injection after half an hour AICAR slaughtered; Adopt western-style imprinting analysis of total protein and in skeletal muscle of the beta 2 AMPK nucleoprotein expression level; And the determination of copper with sulfuric acid in oxygenated muscle glycogen storage changes.Results: After three days load training, the success of the simulated muscle glycogen supercompensation and low muscle glycogen model,And in this model, Skeletal muscle cells in total protein expression of AMPK beta 2 in all groups are no changed,however the AMPK beta 2 expression In nucleoprotein, LE0 group (low muscle glycogen group) is higer than HE0group ( high muscle glycogen group), and muscle concentrations of glycogen significantly reduced; LE group compared to LE0 group, HEgroup compared to HE0 group ,the expresstion of AMPK beta2 in skeletal muscle nucleus protein is higher, muscle glycogen concentrationdecreased.Conclusions: (1) Once trainning, the concentration of muscle glycogen havn't effction with the express of AMPKbeta2 in sketal muscle cell;(2) In skeletal muscle cells ,the positioning of AMPKbeta2 correspon to the change of concentration of muscle glycogen,low muscle glycogen more conducive to the transposition of AMPKbeta2 , high muscle sugar against the transposition of AMPK beta 2 .
