Objective to simulate the fracture healing process of rabbits with femoral fracture and traumatic brain injury model in the rats'simple farcture modles , after injecting serum of the rabbits with fractures and traumatic brain injuries. To analyze this kind of fracture healing mechanism and reveal the clinical applying value.Mehtods 8 rabbits with femoral fracture and traumatic brain injury model were prepared, and then the serum of the 8 rabbits (experimental serum) was prepared, lyophilized and saved at 1w, 2w, 3w, 4w, postoperatively. Negative control serum of 2 healthy rabbits was also prepared, lyophilized and saved. 72 rats with femoral shaft fractures (fixed with Kirschner wire intramedullary) were randomly divided into 3 groups: the experimental group, negative control group and blank control groups. The homologous interval experimental serum were intramuscular injected into the rats of the experimental group at 1d (week 1), 8 days (week 2), 15 days (week 3), 22 days (week 4) after surgery, while negative control serum were intramuscular injected into the rats of the negative control group correspondingly. The serum dose was calculated according to the superficial area rate of the rats and rabbits. The rats of the blank control group had not any management. 6 rats of each group were sacrificed at 1w, 2w, 3w, 4w postoperatively. The effect of experimental serum on the fracture healing of the rats will be analyzed and observed by x-ray, hematoxylin and eosin stain, immunohistochemical staining and blood test by immunneoenzymatic techniques.Results1 x-ray:The callus formation can not be found apparently until 2w after operation in these groups. And the callus formation were more rapid and callus were more dense in experimental group than in the control groups at 2w, 3w postoperatively. What is more, the experimental group have higher gray values of callus(p>0.05) than others at 2w, 3w, 4w in the Picture Archiving and Communication System(VeeYooPACS) .2 hematoxylin and eosin stain:Experimental group: At 1w, a great of granulation tissue, connective tissue, blood capillary fibroblast, secretory collagen fibers and cartilage cell were found in callus .There was no newly bone trabecula. At 2w,the newly bone trabecula formed.At 3w,the newly bone trabecula bigan to connect and shape.At 4w, the bone trabecula start transforming into mosaic of bone.Negative control group and blank control groups : At 1w, a few of granulation tissue, connective tissue, blood capillary fibroblast, secretory collagen fibers and cartilage cell were found in callus .There was no newly bone trabecula. At 2w,the newly bone trabecula formed.At 3w,the newly bone trabecula began to connect, but do not shape.At 4w, the bone trabecula start to shape rather than transform to mosaic of bone.3 immunohistochemical staining (IHCS) :The mean HIS (immunohistochemistrys core) of VEGF(vascular endothelial growth factor) in callus of the fracture experimental group was higher than that of the negative control group and blank control groups at 1w, 2w, 3w, 4w, postoperatively (P<0.05).4 blood test by immunneoenzymatic techniques4.1 The concentration of NGF(nerve growth factor) in serum of experimental group was higher than that of the negative control group and blank control groups at various intervals(P<0.05).4.2 The concentration of TGF-β(transforming growth factor-β)in serum of experimental group was higher than that of the negative control group and blank control groups at various intervals(P<0.05) Conclusion1.The fracture healing environment of the rats with femoral fracture and traumatic brain injury can be simulated by injecting serum of the rabbits with fractures and traumatic brain injuries into rats'simple farcture modles.2 The mechanism of accelerated fracture healing may be attributed to the high expression of VEGF in callus and NGF,TGF-βin serum.3. This research results, have important directive to promoting the fracture healing clinically.
